长链非编码RNA lnc-Cox2在类风湿性关节炎发病中的作用

摘　　要：目的 观察长链非编码RNA lnc-Cox2对成纤维细胞样滑膜细胞(FLS)增殖、凋亡的影响,并探讨其诱发类风湿关节炎(RA)的发病机制。方法 首先采用实时荧光聚合酶链反应法(RT-PCR)检测RA患者和健康志愿者血浆中lnc-Cox2和TNF-α水平;在RA-FLS细胞中抑制lnc-Cox2表达后,通过RT-PCR和Western blot检测TNF-α的表达水平;在RA-FLS细胞中抑制lnc-Cox2表达后,再过表达TNF-α,应用CCK-8法检测转染后各组吸光值以评价增殖率,应用Annexin V-APC单染色流式细胞技术检测各组转染48 h后的细胞凋亡率。结果 与健康人群相比RA患者血浆中lnc-Cox2、TNF-α过表达,两者表达呈正相关;抑制lnc-Cox2表达后RA-FLS细胞中TNF-α表达下调;RA-FLS细胞抑制lnc-Cox2表达后细胞增殖转移能力明显减弱,凋亡增加,而过表达TNF-α后上述表型明显恢复。结论 lnc-Cox2促进TNF-α的表达而调节RA-FLS细胞的增殖、凋亡和化疗耐药,从而参与RA的发生发展。Objective To study the role and mechanism of long noncoding RNA lnc⁃Cox2 in proliferation and apoptosis of fibroblast⁃like synoviocytes cell.Methods The expressions of lnc⁃Cox2 and TNF⁃αin plasma of RA patients and healthy volunteers were detected by real⁃time PCR.The expression of TNF⁃αin RA⁃FLS cells after knockdown lnc⁃Cox2 was examined by RT⁃PCR and Western blot.The proliferationability in RA⁃FLS cells after knockdown lnc⁃Cox2 and overexpressed TNF⁃αwas examined by CCK⁃8.Annexin V⁃APC staining was used to detect the apoptotic rates at 48 h after transfection.Results The expression levels of lnc⁃Cox2 and TNF⁃αin RA⁃plasma were significantly increased as compared with those in healthy volunteers.Transfection with si⁃lnc⁃Cox2 down⁃regulated TNF⁃αexpression.In RA⁃FLS cells,the abilities of proliferation and apoptosis were significantly reduced after knockdown of lnc⁃Cox2.And the overexpression of TNF⁃αcan reversed this phenomenon.Conclusion lnc⁃Cox2 regulated the proliferation and apoptosis of RA⁃FLS cells via promoting the expression of TNF⁃α,and involved in the occurrence and development of RA.
