摘  要:[目的]沙棘是典型的放线菌结瘤植物,是三北地区主要的造林先锋树种。然而,学界对沙棘整体微生物组的群落结构和结瘤固氮共生模式知之甚少。本研究通过分析不同物种和区室生态位对沙棘微生物组结构和组成的影响,系统解析了沙棘微生物组的特性。[方法]采用16S rRNA测序方法,研究了三北地区栽培面积最大的蒙古沙棘和中国沙棘区室生态位微生物群落的多样性和组成结构的差异。[结果]研究发现蒙古沙棘和中国沙棘不同区室微生物群落的多样性和组成结构主要受区室生态位的影响,其中土壤区室细菌的Alpha多样性(Shannon指数在8.50~8.96之间)显著高于植物内生区室(Shannon指数在4.79~8.32之间),而根瘤内生区室的Alpha多样性最低(Shannon指数在4.79~5.69之间)。不同区室微生物群落的细菌组成具有明显差异,其中假黄色单胞菌属和根瘤菌属主要在根内生区室中富集,马赛菌属则主要富集在茎内生和叶内生区室中,而弗兰克氏菌属则在根瘤内生区室中占主导地位,且属于两个沙棘物种根瘤内生区室中共同且唯一的生物标志物。溯源结果表明,沙棘根内生和根瘤内生区室的微生物群落主要来自土壤环境并逐步被过滤和富集,而茎内生和叶内生区室中的微生物群落来源并不是土壤。[结论]区室生态位是影响沙棘微生物群落组成结构和共生模式的主要原因。根瘤作为独特的生态位,Frankia在根瘤中占主导地位,表明沙棘-Frankia的专一性共生关系。这些发现为深入理解沙棘对环境的适应机制提供了重要理论基础。[Objective]Sea buckthorn(Hippophae rhamnoides L.)is a typical actinorhizal plant and a main pioneer afforestation species in the Three-North Region.However,little is known about the overall microbiome community structure and nodule nitrogen fixation symbiosis pattern of sea buckthorn.This study systematically analyzed the microbiome characteristics of sea buckthorn by analyzing the effects of different species and compartment niches on the microbiome structure and composition of sea buckthorn.[Method]We employed 16S rRNA gene amplicon sequencing method to study the diversity and composition of microbial communities in the compartment niches of H.rhamnoides subsp.sinensis and H.rhamnoides subsp.mongonica,which have the largest cultivated area in the Three-North Region.[Result]It was found that the diversity and composition structure of microbial communities of H.rhamnoides subsp.sinensis and H.rhamnoides subsp.mongonica were mainly influenced by the compartment niche.The alpha diversity of bacterial communities in the soil compartments(Shannon index ranged from 8.50 to 8.96)was significantly higher than that of plant endosphere compartment niches(Shannon index ranged from 4.79 to 8.32),with the nodule endosphere compartment showing the lowest alpha diversity(Shannon index ranged from 4.79 to 5.69).Significant differences were observed in the composition of microbial communities across different compartments.Pseudoxanthomonas and Rhizobium were mainly enriched in the root endosphere microbial communities,while Massilia was mainly enriched in the stem endosphere and leaf endosphere compartments.Frankia was dominant in the nodule endosphere compartment and was the sole biomarker for this compartment in both sea buckthorn species.Source tracking analyses indicated that the microbial communities in the root and nodule endosphere compartments were mainly derived from the soils and underwent gradual enrichment and filtering.However,the main microbial sources in the stem and leaf endosphere compartments was not soil-deriv
