芦笋EXP基因家族鉴定及其在茎伸长阶段表达分析

摘  要:【目的】全基因组鉴定芦笋EXP基因家族成员,探究EXP基因参与芦笋嫩茎生长的机制。【方法】利用生物信息学方法对芦笋基因组中的EXP基因家族成员进行鉴定和分析,并通过实时荧光定量PCR验证分析AoEXPs在不同生长阶段的表达情况。【结果】芦笋基因组中共有23个EXP基因成员,随机分布在8个染色体上,编码蛋白均定位于细胞壁。结构和进化分析显示,23个AoEXPs基因可分为EXPA、EXPB及EXPLB 3个亚家族。所有基因均具有3个共同的motif结构,且启动子区域均含有数量不等的与植物激素、细胞分裂伸长相关的顺式作用元件。转录组分析表明,在23个EXP基因成员中,17个EXP在芦笋茎伸长过程中表达,其中10个EXP成员在不同发育阶段呈现显著差异表达特征。进一步采用qRT-PCR验证发现,AoEXPs基因具有各自特异的时空表达模式,并与转录组表达丰度相一致。例如AoEXPA5、AoEXPA7、AoEXPA10和AoEXPB1这4个基因随着嫩茎的伸长,表达水平逐渐降低。进而可以推测这10个EXP基因成员可能参与了调控芦笋的伸长生长的过程。【结论】初步鉴定并提供了芦笋EXP基因家族成员信息,为今后进一步探索芦笋EXP基因功能提供了重要的研究基础。【Objective】To perform a genome-wide identification of the EXP gene family members in asparagus and investigate the mechanisms of EXP genes involvement involved in the growth of asparagus tender stems.【Method】Bioinformatics methods were used to identify and analyze the EXP gene family members in the asparagus genome,and quantitative real-time PCR(qRT-PCR)was utilized to validate the expression patterns of AoEXPs at different growth stages.【Result】The asparagus genome comprises 23 EXP gene members,which are randomly distributed across eight chromosomes,with all encoded proteins localized to the cell wall.Structural and evolutionary analysis showed that these 23 AoEXPs can be classified into three subfamilies:EXPA,EXPB,and EXPLB.All genes possess three common motif structures,and their promoter regions contain varying numbers of cis acting elements related to plant hormones and cell division elongation.The transcriptome data revealed that out of 23 EXP genes,17 Ao⁃EXPs were expressed during asparagus stem elongation,with 10 EXP members were differentially expressed at different developmental stages.qRT-PCR validation revealed that the AoEXPs genes had their own specific spatio-temporal expression patterns,which are consistent with the transcriptome expression abundance.For example,the four genes AoEXPA5,AoEXPA7,AoEXPA10 and AoEXPB1 showed a gradual decrease in expression levels with stem elongation.It was hypothesized that these 10 genes may be involved in the process of regulating the elongation and growth of asparagus stems.【Conclusion】This study preliminarily identified and characterized on the members of the asparagus EXP gene family,laying an important foundation for future research on the functional exploration of AoEXP genes.
