‘黑果枸杞’杂交F1代群体遗传结构及其农艺性状关联分析

摘  要:【目的】挖掘‘黑果枸杞’重要农艺性状相关联的分子标记,为其新品种选育与种质创新提供参考依据。【方法】以‘黑果枸杞’杂交F1代227份群体样本为实验材料,在17个重要农艺性状多样性、相关性及主成分分析的基础上,结合19对SSR引物进行的遗传多样性和遗传结构分析结果,采用Tassel 3.0软件中MLM模型,进行农艺性状与SSR标记的关联分析,筛选标记用于新品种选育。【结果】227份‘黑果枸杞’杂交F1代群体17个表型性状变异系数在20%以上,Shannon指数H'均大于6.5;相关性分析表明叶面积与叶片宽度、叶片长度呈极显著正相关(P<0.01),未成熟果实数与果柄长呈极显著正相关(P<0.01);PCA分析筛选出累计贡献率为88.78%的6个主成分,其中叶片宽度(-0.79)、叶片长度(-0.76)、果实纵径(0.75)、果实横径(0.73)、单株结果主枝条数(0.72)、枝条基部粗度(0.63)、平均单果质量(0.61)、枝条长度(0.57)和花色苷含量(0.41)9个性状为关键性状;多态性信息含量(PIC)平均值为0.758,群体总遗传分化指数(F_(st))为0.010(P<0.05)产生的遗传分化均来自于杂交群体内;遗传结构与聚类分析明确将参试样本分成6个亚群;关联分析(MLM模型)得出5个与叶片宽度、叶片长度、叶片面积等生长性状高度关联的SSR标记,表型变异解释率均在10.00%以上,且存在同一标记与多个性状、同一性状与多个标记高度关联的现象。【结论】‘黑果枸杞’枝条、叶片及果实性状上存在广泛的遗传多样性,5个标记存在“一因多效”现象。【Objective】This study aimed to identify molecular markers related to important agronomic traits in Lycium ruthenicum Murr.to provide a reference for breeding new varieties and germplasm innovation.【Method】A hybrid F1 population of 227 L.ruthenicum samples was used as experimental materials.Based on the diversity,correlation and principal component analysis of 17 important agronomic traits,combined with genetic diversity and population structure analysis using 19 SSR primers.The MLM model in Tassel 3.0 software was used to analyze the association between arterial traits and SSR markers,and the markers were selected for new variety breeding.【Result】The variation coefficients of 17 phenotypic traits in 227 F1 hybrids exhibited a coefficients of variation(CV)exceeding 20%,with Shannon's diversity index(H')all above 6.5.Correlation analysis showed that the leaf area was significantly positively correlated with leaf width and leaf length(P<0.01),while the number of immature fruits was significantly positively correlated with fruit stalk length(P<0.01).PCA identified six principal components with a cumulative contribution rate of 88.78%,among which nine key traits were determined:leaf width(-0.79),leaf length(-0.76),fruit longitudinal diameter(0.75),fruit transverse diameter(0.73),number of main branches per plant fruit(0.72),branch base diameter(0.63),average fruit weight per fruit(0.61),branch length(0.57),anthocyanin content(0.41).Polymorphism information content(PIC)with a mean of 0.758,and the total genetic differentiation index(F_(st))was 0.010(P<0.05),indicating that genetic variation primarily originated within the hybrid population.Genetic structure and clustering analysis divided the samples into six subgroups.Five SSR markers were highly correlated with leaf width,leaf length,leaf area and other growth traits by association analysis(MLM model),and the explanation rate of phenotypic variation was over 10.00%.Notably,same markers were associated with multiple traits,and same traits were linked to mu
