高效液相色谱同时测定甘氨酸和二酮哌嗪含量

摘　　要：建立了高效液相色谱检测甘氨酸和二酮哌嗪含量的方法。确定了色谱条件:色谱柱为Waters Anionic Polar Pesticide(5μm,2.1 mm×100 mm),流动相为甲醇/水(体积比7∶3),加入四丁基溴化铵0.50 g/L、磷酸二氢钾0.27 g/L并加入磷酸调节pH=5.0,检测波长195 nm,柱温30℃,流速0.5 mL/min,进样量20μL。线性相关系数达到0.999以上,样品甘氨酸和二酮哌嗪测定标准偏差分别为0.54%和0.44%,加标回收率91.3%~106.5%。该方法方便快捷,准确度高,重复性好。A method for the determination of glycine and diketopiperazine by HPLC was established.The optimal chromatographic conditions were determined as follows:the column was Waters Anionic Polar Pesticide(5μm,2.1 mm×100 mm),the mobile phase was methanol/water solution(volume ratio 7∶3),with 0.50 g/L tetran-butylammonium bromide and 0.27 g/L potassium dihydrogen phosphate added,with the pH of 5.0 adjusted with phosphoric acid,the detection wavelength was 195 nm.The column temperature was 30℃,the flow rate was 0.5 mL/min,and the injection volume was 20μL.The linear correlation coefficient was over 0.999,and the relative standard deviations(RSD)were 0.54%and 0.44%,and the average recoveries were 91.3%~106.5%.The method is convenient,fast,and has good accuracy and repeatability.
