摘 要:目的HPLC测定9种化学成分含量并建立指纹图谱,结合化学模式识别法对26批地骨皮饮片质量进行评价。方法色谱柱Zorbax C_(18)SB-AQ(250 mm×4.6 mm,5μm),流动相乙腈-0.1%三氟乙酸水溶液,进样体积10μL,柱温40℃,梯度洗脱,流速1.0 mL/min,检测波长280 nm。对地骨皮进行定量,采用聚类分析、主成分分析并计算综合得分进行排名,评价26批地骨皮饮片质量。结果建立了地骨皮饮片HPLC指纹图谱,相似度为0.958~0.998;聚类分析将饮片聚成四大类;主成分分析结果表明样品S21得分最高。结论研究建立了多指标含量测定及指纹图谱结合化学模式识别的方法,为临床中地骨皮饮片的质量评价和合理应用提供了参考。Objective To determine the contents of nine chemical components in Cortex Lycii(地骨皮)using HPLC,establish fingerprint profiles,and evaluate the quality of 26 batches of Lycii Cortex slices(地骨皮饮片)by combining chemical pattern identification.Methods The chromatography was performed on a Zorbax C_(18)SB-AQ column(250 mm×4.6 mm,5μm)with a mobile phase of acetonitrile-0.1%trifluoroacetic acid aqueous solution under gradient elution.The flow rate was 1.0 mL/min,column temperature was 40℃,injection volume was 10μL,and detection wavelength was set at 280 nm.Quantitative analysis of Cortex Lycii components was conducted.Cluster analysis(CA)and principal component analysis(PCA)were applied to calculate comprehensive scores for ranking,thereby evaluating the quality of the 26 batches.Results HPLC fingerprint profiles of Lycii Cortex slices were established with similarity values ranging from 0.958 to 0.998.CA classified the slices into four major groups.PCA results indicated that sample S21 achieved the highest comprehensive scoreConclusion A method combined with multi-component quantitative analysis and fingerprint profiling with chemical pattern identification was successfully established.This approach provides a reference for quality evaluation and rational application of Lycii Cortex slices in clinical practice.
