摘 要:目的比较加味沙参麦冬汤配方颗粒(JWSSMD-k)与加味沙参麦冬汤汤剂(JWSSMD-t)物质成分的异同性。方法建立加味沙参麦冬汤UPLC-ESI-QTOF-MS/MS分析方法,结合质谱碎片、对照品、文献进行化学成分鉴定;采用PCA、OPLS-DA等多元统计分析技术对比JWSSMD-k与JWSSMD-k的异同性,筛选出影响较大指标建立含量测定方法,并比较含量的差异。结果共鉴定了36个共有峰,JWSSMD-k与JWSSMD-t正、负离子模式下总离子流图谱一致性高,化学成分差异较小;PCA不能明显区分二者差异,进一步结合OPLS-DA和t统计分析,29个成分无统计学差异,筛选出了7个差异性成分,建立了差异性成分含量测定方法,该方法稳定性良好;JWSSMD-k组中柯里拉京、没食子酸甲酯、诃子次酸、triglloylglucose、矮地茶苷J等5个成分明显高于JWSSMD-t组,而岩白菜素、苦杏仁苷则低于JWSSMD-t组。结论JWSSMD-k与JWSSMD-t物质成分整体差异性较小,二者具有整体质量一致性与特定成分差异性的特征,该研究为加味沙参麦冬汤配方颗粒临床应用提供依据。Objective To compare the similarities and differences in the material components between Jiawei Shashen Maidong Decoction(加味沙参麦冬汤,JWSSMD)in form of granule(JWSSMD-k)and its decoction(JWSSMD-t).Methods UPLC-ESI-QTOF-MS/MS analytical method was established for JWSSMD.Chemical composition was identified by combining mass spectral fragmentation,reference substances and literature.Multivariate statistical analysis techniques,including PCA and OPLS-DA,were employed to compare the similarities and differences between JWSSMD-k and JWSSMD-t.High-impact influential components were screened to establish a quantitative determination method,and content differences were compared.Results A total of 36 common peaks were identified.The total ion chromatograms of JWSSMD-k and JWSSMD-t under positive and negative ion modes showed high consistency,with minor differences in chemical components.PCA could not distinctly differentiate between the two,but further OPLS-DA and t-statistical analyses revealed no statistically differences in 29 components.Seven differential components were identified,and a stable quantitative method of differential ingredient content was established.In the JWSSMD-k group,five components(corilagin,methyl gallate,chebulic acid,triglloylglucose,and ardisiacrispin J)were significantly higher than those in JWSSMD-t groups,while bergenin and amygdalin was lower.Conclusion The overall differences in material components between JWSSMD-k and JWSSMD-t are insignificant,demonstrating both overall quality consistency and specific component variations.This study provides a basis for the clinical application of JWSSMD-k.
