摘 要:目的获得区分芍药和川赤芍的方法,为不同基源赤芍的鉴别提供理论依据。方法使用数码体式显微镜、数码显微镜观察并拍摄芍药及川赤芍根表面及横切面的特征,鉴定两者区别;使用70%乙醇为溶剂对不同基源赤芍中化学成分进行提取,采用UPLC-Q-Exactive-MS/MS技术对其成分进行解析。结果显微鉴定得出川赤芍比芍药横切面有更为紧密的木部放射状纹理,有更为紧密的射线细胞。UPLC-Q-Exactive-MS/MS技术得出,葡萄糖酸、(-)-没食子酸儿茶素酯、表儿茶素没食子酸酯、4-甲基-7-氧香豆素-β-D-葡萄糖苷酸、牡丹皮苷C、儿茶素共计6个化合物仅在芍药中被检出;间甲酚、1,6-脱水-2,4-O-苯基-B-D-吡喃葡萄糖、D-(-)-水杨苷、银杏内酯C、Ginnalin A、4-甲基-7-氧香豆素-β-D-葡萄糖苷酸、Corchorifatty acid F、地诺前列素氨丁三醇、2,2'-亚甲基双(4-甲基-6-三级丁基酚)共计9个化合物仅在川赤芍中检出。结论采用显微镜可初步判别芍药及川赤芍,且两者化学活性成分有明显区别,该研究为不同基源赤芍鉴别方法提供了科学的理论依据。Objective To obtain a method for distinguishing Paeonia lactiflora Pall.(P.lactiflora)and Paeonia veitchii Lynch(P.veitchii),providing a theoretical basis for the identification of Paeoniae Rubra Radix(PRR)from different origins.Methods The surface and transverse section characteristics of P.lactiflora and P.veitchii roots were observed and photographed using digital stereomicroscopy and digital microscopy to identify their differences.Chemical components of PRR from different origins were extracted using 70%ethanol as the solvent and analyzed by UPLC-Q-Exactive-MS/MS.Results Microscopic identification revealed that P.veitchii exhibits a denser radial arrangement of xylem and more tightly packed ray cells in its transverse section compared to P.lactiflora.According to the results obtained by UPLC-Q-Exactive-MS/MS technology,a total of six compounds:gluconic acid,(-)-gallocatechin gallate,epicatechin gallate,4-methyl-7-oxycoumarin-β-D-glucuronide,mudanpioside C,and catechin were detected exclusively in P.lactiflora.Nine compounds including m-cresol,1,6-anhydro-2,4-O-phenyl-B-D-glucopyranose,D-(-)-salicin,ginkgolide C,ginnalin A,4-methyl-7-oxycoumarin-β-D-glucuronide,corchorifatty acid F,dinoprost tromethamine,and 2,2'-methylenebis(4-methyl-6-tert-butylphenol)were uniquely identified in P.veitchii.Conclusion Microscopy can preliminarily distinguish P.lactiflora and P.veitchii,and their chemical active components exhibit significant differences.This study provides a scientific theoretical basis for the identification of PRR from different origins.
