摘 要:目的观察秩边透水道针法对原发性痛经(PD)大鼠下丘脑-垂体-卵巢(HPO)轴功能的影响。方法将雌鼠随机分为空白组、模型组和针刺组(n=10)。模型组和针刺组采用苯甲酸雌二醇联合缩宫素构建PD大鼠模型,针刺组给予秩边透水道针法干预。第11天注射缩宫素后观察各组大鼠扭体潜伏期及次数,计算评分;HE染色观察子宫组织病理改变;ELISA法检测大鼠血清中PGF_(2α)、GnRH、FSH、LH、E_(2)、P含量;Western blot检测垂体组织中GnRH-R、FSHR、LHR、ER、PR蛋白表达量;RT-qPCR法检测垂体组织中GnRH-R、卵巢组织中FSHR、LHR、子宫组织中ER、PR mRNA表达水平。结果与空白组比较,模型组大鼠出现扭体反应,扭体潜伏期缩短(P<0.01),扭体次数和评分增高(P<0.01),大鼠子宫内膜剥落并伴严重水肿及炎性细胞浸润,血清中PGF_(2α)、GnRH、FSH、LH、E_(2)含量增加(P<0.01),血清中P含量减少(P<0.01),垂体组织中GnRH-R、卵巢组织中FSHR、LHR、子宫组织中ER蛋白和mRNA表达升高(P<0.01),PR蛋白和mRNA表达降低(P<0.01);与模型组比较,针刺组大鼠扭体潜伏期延长(P<0.01),扭体次数和评分下降(P<0.01),大鼠子宫损伤改善,血清中PGF_(2α)、GnRH、FSH、LH、E_(2)含量减少(P<0.01,P<0.05),血清中P含量增加(P<0.01),垂体组织中GnRH-R、卵巢组织中FSHR、LHR、子宫组织中ER蛋白和mRNA表达降低(P<0.05,P<0.01),PR蛋白和mRNA表达升高(P<0.01)。结论秩边透水道针法可能是通过调节HPO轴相关激素及其受体蛋白和基因表达水平,改善PD大鼠内分泌紊乱,并减轻疼痛症状。Objective To observe the effects of Zhibian(BL 54)-to-Shuidao(ST 28)(ZBTSD)acupuncture on the function of the hypothalamic-pituitary-ovarian(HPO)axis in rats with primary dysmenorrhea(PD).Methods Female rats were randomly divided into the blank group,model group,and acupuncture group(n=10).The model and acupuncture groups were established as PD rat models using estradiol benzoate combined with oxytocin.The acupuncture group received intervention of ZBTSD acupuncture.On day 11,after oxytocin injection,writhing latency,frequency,and scores were observed.Uterine histopathology was assessed via HE staining.ELISA was used to measure the contents of PGF2α,GnRH,FSH,LH,E2,and P in serum.Western blot(WB)was performed to detect protein expressions of GnRH-R,FSHR,LHR,ER,and PR in the pituitary tissues.RT-qPCR was used to measure mRNA expression levels of GnRH-R in the pituitary,FSHR and LHR in the ovaries,and ER and PR in the uterus.Results Compared with the blank group,the model group exhibited writhing responses,shortened writhing latency(P<0.01),increased writhing frequency and scores(P<0.01),endometrial shedding with severe edema and inflammatory cell infiltration,elevated levels of PGF2α,GnRH,FSH,LH,and E2 in serum(P<0.01),decreased serum P(P<0.01),up-regulated protein and mRNA expression of GnRH-R in the pituitary,FSHR and LHR in the ovaries,and ER in the uterus(P<0.01),and down-regulated PR protein and mRNA expression(P<0.01).Compared with the model group,the acupuncture group showed prolonged writhing latency(P<0.01),reduced writhing frequency and scores(P<0.01),alleviated uterine damage,decreased contents of PGF2α,GnRH,FSH,LH,and E2 in serum(P<0.01,P<0.05),increased serum P contents(P<0.01),down-regulated expression levels of GnRH-R in the pituitary tissues,FSHR and LHR in the ovarian tissues,and ER protein and mRNA in the uterine tissues.Conclusion ZBTSD acupuncture may alleviate endocrine disorders and pain symptoms in PD rats by regulating HPO axis-related hormones and their receptor protein and gene expressi
