摘  要:以无侧枝切花菊品种‘圣雪’为试材,以茎尖和单芽茎段2种材料为外植体,采用植物组织培养的方法,研究了不同外植体、不同浓度植物生长调节剂对切花菊生根和分化的影响,以期建立高效稳定的无侧枝切花菊组织培养再生体系,为解决我国市场中缺乏无侧枝切花菊种苗提供参考依据。结果表明:茎尖比单芽茎段更适合作为外植体,其中4~5 mm的茎尖效果比2~3 mm的茎尖好。在茎尖的初代培养中,MS+0 mg·L^(-1)6-BA+0 mg·L^(-1)NAA更适宜用来直接促进茎尖萌发,MS+2.0 mg·L^(-1)6-BA+0.3 mg·L^(-1)NAA与MS+1.0 mg·L^(-1)6-BA+0.1 mg·L^(-1)NAA更适宜用来诱导丛生芽产生,MS+1.0 mg·L^(-1)6-BA+0.1 mg·L^(-1)NAA和1/2MS+0.1 mg·L^(-1)NAA分别可以用作菊花的继代与生根培养基使用,增殖、生根效果均良好。最终建立了切花菊品种‘圣雪’高效稳定的组织培养再生体系。Taking the stem tips and single bud stem segments of the cut chrysanthemum variety‘Shengxue’as the test material,the effects of different materials and concentrations of plant growth regulators on the rooting and differentiation of cut chrysanthemum were studied by using plant tissue culture techniques,in order to establish a highly efficient and stable tissue culture and regeneration system for cut chrysanthemum without lateral branch,and provide reference for solve the lack of lateral branch cut chrysanthemum varieties in the Chinese market.The results showed that the stem tip was more suitable as an explant than a single bud stem segment,with a 4-5 mm shoot tip having better results than a 2-3 mm stem tip.In the initial culture of stem tips,MS+0 mg·L^(-1)6-BA+0 mg·L^(-1) NAA was the optimal medium.MS+1.0 mg·L^(-1)6-BA+0.1 mg·L^(-1) NAA and 1/2MS+0.1 mg·L^(-1) NAA could be used as subculture and rooting media for chrysanthemum,respectively,the effects of proliferation and rooting were better.Finally,an efficient and stable tissue culture regeneration system for‘Shengxue’has been developed.
