摘 要:以林下参种子为试材,采用组培快繁的方法,研究了不同消毒时间、基本培养基类型以及光照条件等因素对种子无菌萌发的影响,以期为林下参种子无菌萌发体系的建立及离体快繁技术的应用提供参考依据。结果表明:林下参种子最适消毒处理为使用消毒剂2%NaClO对种子表面进行消毒10 min,然后对种胚进行消毒7 s,该条件下种子消毒效果与萌发效果均为最佳;MS培养基为种子无菌萌发最适基本培养基,最适光照强度为1500 lx;添加单一激素6-BA或NAA可明显提高种子萌发率。整体萌发生长情况最佳的培养基为MS+6-BA 1.2 mg·L^(-1)+NAA 0.3 mg·L^(-1)。The seeds of understory Panax ginseng were used as the experimental materials,tissue culture and rapid propagation was adopted,the effects of different disinfection time,basic medium type and light conditions on aseptic germination of seeds were studied,in order to provide reference for the establishment of aseptic germination system and the application of the rapid propagation technology in vitro of understory Panax ginseng.The results showed that the most suitable disinfection treatment for the seeds of understory Panax ginseng was to disinfect the seed surface with 2%NaClO for 10 minutes,and then disinfect the embryo for 7 seconds,under these conditions,the seed disinfection and germination effects were both optimal.MS medium was the most suitable basic medium for aseptic germination of seeds,and the optimum light intensity was 1500 lx.The addition of a single hormone 6-BA or NAA could significantly improve the seed germination rate.The best medium for the overall germination and growth was MS+6-BA 1.2 mg·L^(-1)+NAA 0.3 mg·L^(-1).
