摘 要:呼吸道合胞病毒(Respiratory syncytial virus,RSV)是引起婴幼儿和儿童急性下呼吸道感染的主要病原体之一,严重时会造成死亡,对人类和社会危害极大。本研究旨在探讨针对RSV融合蛋白(F蛋白)的不同表位中和抗体联合使用对RSV的抑制效果,并采用棋盘法进行联合用药实验,分析各抗体组合对病毒的抑制作用。实验结果表明,所有抗体组合的抑制效果均可通过MacSynergy graphs和ZIP分析方法进行评估。不同表位的抗体联合使用时,增强了抗体的抗病毒效果,经数据分析为叠加效应,未发现明显的拮抗或协同效应。本研究为抗体联合使用治疗RSV感染的策略提供了初步数据支持,为优化RSV的治疗方案提供了参考。未来的研究应进一步深入探讨这些抗体及其联合在RSV实际感染模型中的表现,以期获得最佳的临床效果。Respiratory syncytial virus(RSV)is a major pathogen that causes acute lower respiratory tract infections and even death in infants and young children.It seriously threatens public health and leads to heavy economic burden.This study aims to investigate the inhibitory effects against RSV by combining neutralizing antibodies targeting different epitopes of the RSV fusion(F)protein.Using the checkerboard assay,we evaluated the inhibitory effect of the combinations of various neutralizing antibodies against RSV.The results demonstrated that the inhibitory effects of all tested antibody combinations could be assessed using MacSynergy graphs and ZIP analysis methods.Data analysis revealed that the combined use of antibodies targeting different epitopes may only result in additive effects on antiviral activity.No evidence of significant antagonistic or synergistic effects was observed.This study provides preliminary data to support the strategy of combinational use of antibodies for RSV treatment,which could be useful for optimization of treatment approach against RSV infection.Future studies should further explore the performance of these antibodies and their combinations in actual infection model of RSV to achieve optimal clinical efficacy.
