摘 要:目的:本研究通过分析单细胞数据集和bulk数据集,确认结直肠癌中的主要免疫细胞群及其功能特性。方法:将来自GEO数据库的数据集GSE87211、GSE29638、GSE143985和GSE110224分别按照已知免疫元基因分为两个免疫浸润水平组,在高免疫侵润组中发现14个共同差异表达基因。然后,在单细胞数据集GSE132465的37958个细胞中鉴定出8个主要细胞簇并定位标记基因,以确定肿瘤微环境内的主要细胞群。结果:从单细胞数据集GSE132465中鉴定出一组肿瘤相关巨噬细胞,14个差异表达基因几乎全部在该巨噬细胞中高表达。该肿瘤相关巨噬细胞作为结直肠癌的肿瘤微环境的中心调节因子,与上皮细胞、单核细胞等几个关键细胞亚群密切相关,主要通过MIF-(CD74+CXCR4)通路与肿瘤微环境中的其他细胞进行相互作用。结论:结直肠癌恶性上皮细胞主要通过MIF-(CD74+CXCR4)通路促使单核细胞向肿瘤部位募集并分化为特定的肿瘤相关巨噬细胞,从而促进肿瘤生长和发展。This study aimed at identify the major immune cell populations and their functional characteristics in colorectal cancer by analyzing both single-cell and bulk datasets.Datasets GSE87211,GSE29638,GSE143985,and GSE110224 from the GEO database were divided into two immune infiltration levels based on known immune metagenes.Fourteen common differentially expressed genes were identified in the high immune infiltration group.Eight major cell clusters were identified from the 37958 cells in the single-cell dataset GSE132465,and marker genes were located to determine the predominant cell populations within the tumor microenvironment.A cluster of tumor-associated macrophages was identified from the single-cell dataset GSE132465,with almost all the 14 differentially expressed genes highly expressed in these macrophages.These tumor-associated macrophages act as the central regulatory factors in the tumor microenvironment of colorectal cancer,closely associated with several key cell subgroups such as epithelial cells and monocytes,primarily interacting with other cells in the tumor microenvironment through the MIF-(CD74+CXCR4)pathway.In conclusion,malignant epithelial cells in colorectal cancer predominantly recruit and differentiate monocytes into specific tumor-associated macrophages through the MIF-(CD74+CXCR4)pathway,thereby promoting tumor growth and development.
