参葛补肾胶囊中太子参的薄层色谱鉴别研究

摘  要:目的:确定参葛补肾胶囊中太子参的最佳薄层色谱条件,以建立可操作、专属性强、准确可靠的质量控制方法,为其质量标准修订提供参考。方法:以太子参对照药材为指标成分,将参葛补肾胶囊提取液在层析板上点样,考察展开剂、显色剂对薄层色谱分析的影响,以筛选出最佳的薄层色谱条件。结果:采用硅胶G薄层板,以75%异丙醇-无水乙醇(9∶1)为展开剂,喷以1%~2%的苯胺丙酮溶液、1%~2%的二苯胺丙酮溶液和磷酸按照体积比为(4~5)∶(3~5)∶1组成的混合溶液,在105℃加热至斑点显色清晰,置日光下检视。该色谱斑点清晰,各组分分离效果好,且在与对照药材色谱相应的位置上,均显相同颜色的斑点。结论:该方法安全、易操作、专属性强、重复性良好,可用于参葛补肾胶囊中太子参的定性鉴别。In this study we determined the optimal TLC conditions for Pseudostellaria heterophylla in Shenge Bushen Capsule,so as to establish an operable,specific,accurate and reliable quality control method and provide reference for the quality standard revision of Shenge Bushen Capsule.The extract of Shenge Bushen Capsule was spotted on a chromatography plate with the control herb of Pseudostellaria heterophylla as the index component,and the effects of developing agent and chromogenic agent on TLC analysis were investigated to determine the best TLC conditions.The results showed that,when silica gel G thin layer plate was used,75%isopropanol-absolute ethanol(9∶1)was used as the developing solvent,the mixed solution of 1%-2%aniline acetone solution,1%-2%diphenylamine acetone solution and phosphoric acid was sprayed at a volume ratio of(4-5)∶(3-5)∶1,and the plate was heated at 105℃and viewed in sunlight,the chromatographic spots were clear,the separation effect of each component was good,and the spots of the same color were displayed in the corresponding positions of the chromatogram of the control material.This method is safe,easy to operate,and has strong specificity and good reproducibility,which can be used for the qualitative identification of Pseudostellaria heterophylla in Shenge Bushen Capsule.
