摘 要:目的:探讨大鼠去卵巢后促性腺激素释放激素受体(GnRHR)的变化及胎肾细胞悬液(FRCS)对其干预机制。方法:取3月龄未交配健康SD雌性大鼠64只,随机分为四组,即假手术组、模型组、雌激素组、胎肾组。其中假手术组行假手术,模型组、雌激素组、胎肾组行去卵巢手术。术后8周,假手术组、模型组给予0.9%氯化钠溶液,雌激素组给予苯甲酸雌二醇,胎肾组给予胎肾细胞悬液。药物治疗4周后,利用免疫组织化学、RT-PCR检测GnRHR在下丘脑的表达。结果:与假手术组相比,模型组GnRHR表达量明显升高(均P<0.01);与模型组相比,雌激素组、胎肾组GnRHR表达量降低(P<0.05);干预两组间两两比较,差异均无统计学意义(P>0.05)。结论:去卵巢后大鼠GnRHR表达量增高,应用胎肾细胞悬液进行干预,能降低GnRHR表达量,增加机体的雌激素水平,延缓机体绝经年龄。Objective:To observe the change of Gonadotropin releasing hormone receptor(GnRHR)in rats after ovariectomy and the intervention effect of fetal renal cell suspension(FRCS)on them.Methods:Sixty-four 3-month-old unmated healthy SD female rats were taken and randomly divided into four groups:normal group,model group,estrogen group,and fetal renal cell suspension group.Eight weeks after operation,the rats in normal group and model group were treaded with sodium chloride,the estrogen group was treated with estrogen,the fetal renal cell suspension group was treated with FNCS.All of the rats were sacrificed after four weeks.Immunohistochemistry and RT-PCR methods were used to observer the changes in the expression of GnRHR in hypothalamus.Results:GnRHR levels in model group were substantially higher than in normal group(P<0.01).GnRHR levels in estrogen group and fetal renal cell suspension group were considerably lower than in model group(P<0.05),although there was no significant difference between the two groups in paired comparisons(P>0.05).Conclusion:Rats with ovariectomies undoubtedly have high levels of free radicals;FRCS can reduce these damage and improve oxidation-antagonistic effects.
