摘 要:目的:研究醒脑静对阿尔茨海默病(AD)大鼠学习记忆能力及相关蛋白环腺苷酸应答元件结合蛋白(CREB)表达的影响。方法:选用健康雄性18月龄SD大鼠35只,经过筛选剔除5只不具备游泳能力的大鼠。将符合标准的大鼠进行随机分组,分为空白对照组、醒脑静组、模型组,每组10只大鼠。采用Aβ25-35淀粉样蛋白双侧海马区注射诱发大鼠脑内神经元损伤,制备大鼠AD模型;造模后7 d,醒脑静组大鼠按剂量4 mL/d ig每日上午给药,模型组与空白对照组均经ig途径给予等容量生理盐水,持续治疗28 d。采用Morris水迷宫法连续5 d检测大鼠的学习记忆能力。在末次给药后,处死大鼠并采集其脑标本。运用Western blot法检测大鼠海马中CREB蛋白的表达水平。结果:Morris水迷宫测试结果显示,相较于模型组,醒脑静组大鼠在实验的第1~5天逃避潜伏期表现出缩短趋势,穿越平台次数呈现出增加的特点(P<0.01);同时醒脑静组脑组织中CREB蛋白表达水平升高(P<0.01)。结论:醒脑静注射液可以改善Aβ25-35淀粉样蛋白诱导AD模型大鼠的学习记忆能力,提高海马组织内的CREB蛋白水平。Objective:To investigate the effects of Xingnaojing injection on the learning-memory ability and the expression of cAMP-response element binding protein(CREB)in rat models of Alzheimer′s disease(AD).Methods:Thirty-five healthy male SD rats(18-month-old)were initially selected,in which five rats that were unable to swim were excluded via screening.The rest were randomly divided into 3 groups:blank control group,Xingnaojing group,AD model group.Each group contained 10 rats.The AD rat models were developed by inducing neuronal damage in the brain through bilateral hippocampal injection of Aβ25-35 amyloid protein.Seven days after modeling,rats in the Xingnaojing group underwent intragastric administration(ig)of Xinignaojing in daily dose of 4 mL every morning.Rats in group C and A were both given an equal volume of physiological saline via intragastric administration.After 28 consecutive days of treatment,the learning-memory ability of the model rats was assessed using the Morris water maze test for five consecutive days.After the final administration,the rats were sacrificed and the brain samples were taken.Western Blot was used to detect the expression of CREB protein in the hippocampus of rats.Results:The Morris water maze test results showed that,compared to the model group,the rats in the Xingnaojing group exhibited a significant trend of shortened escape latency during the first to the fifth day of the experiment,and demonstrated a notable increase in the number of platform crossings(P<0.01).The expression of CREB protein in brain tissue was significantly increased in the Xingnaojing group(P<0.01).Conclusion:Xingnaojing injection can significantly improve the learning-memory ability of model rats of AD induced by Aβ25-35 amyloid,and increase the level of CREB protein in the hippocampus.
