摘 要:目的借助网络药理学和生物信息学技术预测六味地黄丸防治高血压阴虚证的分子机制。方法利用基因表达综合(GEO)、GeneCards数据库筛选高血压差异表达基因。应用R.3.6.3软件获取高血压与阴虚证交集基因进行基因本体论(GO)功能富集、京都基因与基因组数据库(KEGG)通路富集分析。运用中药系统药理学(TCMSP)数据库建立六味地黄丸活性成分靶点数据集。采用R.3.6.3软件获取高血压阴虚证“病-证”交集靶点与六味地黄丸活性成分潜在靶点交集靶点,应用STRING网站筛选潜在核心靶点。结果高血压与阴虚证33个交集基因中有14个与炎症密切相关。高血压阴虚证与六味地黄丸“病-证-方”8个交集靶点中有5个与炎症高度相关。六味地黄丸可能通过Toll样受体4(TLR4)介导炎症信号通路防治高血压阴虚证。结论六味地黄丸防治高血压阴虚证机制主要集中在炎症方面,TLR4介导炎症信号通路可能是六味地黄丸防治高血压阴虚证核心信号通路。Objective To predict the mechanism of Liuwei Dihuang Pill in preventing and treating hypertension with yin deficiency syndrome based on network pharmacology and bioinformatics technology.Methods Gene expression omnibus(GEO)and GeneCards database were used to screen differentially expressed genes in hypertension.Using R.3.6.3 software,the intersection genes of hypertension with yin deficiency syndrome were obtained for gene ontology(GO)function enrichment and Kyoto gene and genome database(KEGG)pathway enrichment analysis.The target data set of active components of Liuwei Dihuang Pill was established by using TCM systematic pharmacology(TCMSP)database.R.3.6.3 software was used to obtain the intersection target of"diseasesyndrome"of hypertension with yin deficiency syndrome and the potential target of active components of Liuwei Dihuang Pill,and STRING website was used to screen potential core targets.Results 14 of the 33 intersection genes between hypertension and yin deficiency syndrome were closely related to inflammation.5 of the 8 intersecting targets of hypertension with yin deficiency syndrome and Liuwei Dihuang Pill"disease-syndrome-prescription"were highly related to inflammation.Liuwei Dihuang Pill might prevent and treat hypertension with yin deficiency syndrome through Toll-like receptor 4(TLR4)mediated inflammatory signaling pathway.Conclusion The mechanism of Liuwei Dihuang Pill in preventing and treating hypertension with yin deficiency syndrome mainly focuses on inflammation,and TLR4-mediated inflammatory signal pathway may be the core signal pathway of Liuwei Dihuang Pill in preventing and treating hypertension with yin deficiency syndrome.
