摘 要:目的探讨三叶青(TH)不同部位提取物对硫代乙酰胺(TAA)诱导斑马鱼肝损伤模型的保护作用。方法将斑马鱼暴露于TAA中建立肝损伤模型,给予TH不同部位提取物,测定幼鱼脂质累积、肝功能酶活性等指标,考察TH不同部位的保肝作用。实时定量聚合酶链反应(RT-qPCR)测定斑马鱼中B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白(Bax-2)、平滑肌肌动蛋白α(ACTA2)和转化生长因子-β(TGF-β)的mRNA表达水平,探讨其潜在机制。结果与模型组比较,2 mg/ml叶和根提取物、0.05 mg/ml茎提取物可以明显减轻脂质沉积,增加肝脏面积,降低丙氨酸氨基转移酶、天冬氨酸氨基转移酶活性及丙二醛水平。苏木素-伊红染色结果显示,仅2 mg/ml根提取物明显改善斑马鱼肝细胞空泡化现象。RT-qPCR结果显示,2 mg/ml根提取物能下调肝纤维化相关基因ACTA2、TGF-β的mRNA表达量,下调Bax-2基因的表达,上调Bcl-2基因的表达。结论2 mg/ml根提取物对TAA诱导的斑马鱼肝损伤的保护效果最佳,可能与抗纤维化和细胞凋亡相关,为TH药理研究及临床应用等提供科学依据。Objective To investigate the protective effects of extracts from different parts of Tetrastigma hemsleyanum Diels&Gilg(TH)on a liver injury model in zebrafish induced by thioacetamide(TAA).Methods Zebrafish were exposed to TAA to establish a liver injury model,and extracts from different parts of TH were given.The lipid accumulation and hepatic function enzyme activity of juvenile fish were measured to investigate the hepatoprotective effect of TH in different parts.Real time quantitative polymerase chain reaction(RT-qPCR)was used to measure the mRNA expression levels of B-cell lymphoma/leukemia 2(Bcl-2),Bcl-2 associated X protein(Bax-2),α-smooth muscle actin(ACTA2),and transforming growth factor beta(TGF-β)in zebrafish,and their potential mechanisms were investigated.Results Compared with the model group,2 mg/ml leaf and root extracts and 0.05 mg/ml stem extract significantly reduced lipid deposition,increased liver area,decreased alanine aminotransferase and aspartate aminotransferase activity,and malondialdehyde levels.The results of hematoxylin eosin staining showed that only 2 mg/ml root extract significantly improved the vacuolization phenomenon of zebrafish liver cells.The RT-qPCR results showed that 2 mg/ml root extract could downregulate the mRNA expression levels of liver fibrosis related genes ACTA2 and TGF-β,downregulate the expression of Bax-2 gene,and upregulate the expression of Bcl-2 gene.Conclusion 2 mg/ml root extract has the best protective effect on TAA induced zebrafish liver injury,which may be related to anti-fibrosis and cell apoptosis,providing scientific basis for pharmacological research and cli nical application of TH.
