摘 要:本研究旨在利用非靶向代谢组学技术探究罗伊氏乳杆菌I5007对大鼠营养代谢的影响。选取24只无特定病原体(SPF)级SD雄性断奶大鼠,随机分为3组,每组8个重复,每个重复1只。试验A组(单次灌胃组)于第7天灌胃罗伊氏乳杆菌I5007菌液,于第14天、第21天和第28天灌胃生理盐水;试验B组(多次灌胃组)于第7天、第14天、第21天、第28天灌胃罗伊氏乳杆菌I5007菌液;对照组于第7天、第14天、第21天、第28天灌胃生理盐水。试验期为30 d。试验结束后采集门静脉血液分析血清代谢物,采集空肠组织检测免疫球蛋白和细胞因子含量。结果表明:1)试验B组的终末体重极显著高于对照组(P<0.01);试验A组和试验B组的平均日增重显著或极显著高于对照组(P<0.05或P<0.01)。2)试验B组小肠长度显著低于试验A组和对照组(P<0.05);试验A组和试验B组肝脏指数均极显著低于对照组(P<0.01)。3)与对照组相比,试验A组空肠组织中免疫球蛋白M含量极显著提高(P<0.01);试验A组和试验B组空肠组织中白细胞介素-1α含量显著提高(P<0.05)。4)与对照组相比,试验A组共鉴定到31个显著上调代谢物和12个显著下调代谢物;试验B组共鉴定到28个显著上调代谢物和27个显著下调代谢物。罗伊氏乳杆菌I5007影响了甘油磷脂代谢、鞘脂代谢、精氨酸与脯氨酸代谢、泛酸和辅酶A生物合成、柠檬酸循环等代谢通路。综上可知,罗伊氏乳杆菌I5007通过调节大鼠体内营养代谢,尤其是能量和氨基酸代谢,进而调控营养物质沉积,最终影响大鼠的生长速度、器官发育和免疫功能。This study aimed to explore the effects of Lactobacillus reuteri I5007 on nutrient metabolism in rats by using untargeted metabolomics technology.Twenty-four specific pathogen free(SPF)grade SD male weaned rats were randomly divided into 3 groups with 8 replicates per group and 1 rat per replicate.Lactobacillus reuteri I5007 on the 7th day,and normal saline on the 14th,21st and 28th day was given intragastrically in test group A(single gavage group);Lactobacillus reuteri I5007 on the 7th,14th,21st and 28th day was given intragastrically in test group B(multiple gavage group);normal saline on the 7th,14th,21st and 28th day was given intragastrically in control group.the experiment lasted for 30 days.At the end of the experiment,blood from the portal vein was collected for metabolomics analysis,and jejunum tissue was collected to detect the contents of immunoglobulins and cytokines.The results showed as follows:1)the final body weight of test group B was extremely significantly higher than that of the control group(P<0.01);the average daily gain of both test group A and test group B was significantly or extremely significantly higher than that of the control group(P<0.05 or P<0.01).2)The length of the small intestine of test group B was significantly lower than that of the control group and test group A(P<0.05);the liver index of test group A and test group B was extremely significantly lower than that of the control group(P<0.01).3)Compared with the control group,the content of immunoglobulin M in the jejunum tissue of the test group A was extremely significantly increased(P<0.01),and the content of interleukin-1αin the jejunum tissue of test group A and test group B was significantly increased(P<0.05).4)Compared with the control group,a total of 31 significantly up-regulated metabolites and 12 significantly down-regulated metabolites were identified in test group A,and a total of 28 significantly up-regulated metabolites and 27 significantly down-regulated metabolites were identified in test group B.Lactobacillu
