摘 要:目的 从野生冬虫夏草中的核苷类、甾醇类及多糖类成分入手,基于4种核苷(腺苷、鸟苷、肌苷、尿苷)、3种甾醇(麦角甾醇、胆甾醇、谷甾醇)及多糖含量的变化,对不同地区采集的冬虫夏草进行定量分析和综合评价,通过建立模型评价和预测冬虫夏草评级。方法 建立HPLC方法测定产自西藏那曲、西藏山南以及青海玉树的冬虫夏草中4种核苷和3种甾醇的含量,采用苯酚—硫酸法测定虫草粗多糖含量,并结合主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)以及火山图分析等分析方法,寻找并分析不同产地冬虫夏草的成分差异。结果 不同产地冬虫夏草中均含有4种核苷和3种甾醇,不同产地含量存在差异,通过化学模式识别分析结果,并对冬虫夏草的质量评价指标进行筛选,从而对其评级进行预测。结论 本文所开展的多成分含量综合评价可以更全面的对冬虫夏草质量进行分析,可为制订冬虫夏草等级质量标准提供科学的参考依据。Objective Based on the content change of 4 nucleosides(adenosine,guanosine,inosine,uridine),3 sterols(ergosterol,cholesterol,sterol)and polysaccharides in Cordyceps sinensis,the components of Cordyceps sinensis from different origins of medicinal materials were quantitatively analyzed and quality evaluated.A model was established to evaluate and predict the cordyceps sinensis rating.Methods A HPLC method was established to determine the contents of 4 nucleosides and 3 sterols in Cordyceps sinensis from Naqu in Tibet,Shannan in Tibet and Yushu county in Qinghai Province.The content of polysaccharides in Cordyceps sinensis was determined by the phenol-sulfuric acid method.Combined with orthogonal partial least squares and principal component analysis(PCA),multiplying discriminant analysis(OPLS-DA)and volcano map analysis,this article found and analyzed the difference in composition content of Cordyceps sinensis from different origins of medicinal materials.Results Cordyceps sinensis collected from different origins all contained 4 nucleosides,3 sterols and polysaccharides.However,the contents of compounds in different origins were different.The quality evaluation indexes of Cordyceps sinensis were screened through the results of chemical pattern recognition analysis,so as to predict its rating.Conclusion Through multi-component content determination,the quality of Cordyceps sinensis can be evaluated more scientifically and comprehensively,which lays a solid foundation for further analysis of the components of Cordyceps sinensis from different origins,and also provides a basis for the rational use of different grades of Cordyceps.It can also be used as a commercial grade quality standard for Cordyceps sinensis.
