摘 要:α-微管蛋白在棉花纤维细胞伸长和细胞骨架构建中扮演着重要角色。利用生物信息学技术,鉴定了棉花TUA基因家族成员并分析了其组织表达模式,为深入研究TUA基因在棉花纤维发育中的作用提供理论依据。采用MEGA11.0构建系统进化树,绘制染色体定位图,利用MEME分析保守基序,并通过PlantCARE分析启动子顺式作用元件;同时,利用转录组数据分析陆地棉TUA基因的组织表达特性。结果表明:在4个棉种内共含有67个TUA家族成员,且在四倍体棉种中的数目约为二倍体棉种数目的2倍,系统进化将其分为A-E共5个亚组。TUA基因家族在陆地棉与海岛棉之间的进化具有相似性。顺式作用元件分析预测显示大部分元件与生长发育和防御胁迫相关。利用TM-1转录组数据分析陆地棉的23个TUA基因的组织表达特性,发现TUA基因在纤维中优势表达。筛选出陆地棉纤维优势表达的TUA基因家族成员,为棉花纤维性状的遗传改良提供了重要的候选基因资源。Alpha-tubulin plays a crucial role in the elongation of cotton fiber cells and the construction of the cytoskeleton.Using bioinformatics techniques,this study aims to identify members of the TUA gene family in cotton and analyze their tissue expression patterns,to provide a theoretical basis for the in-depth study of the role of TUA genes in cotton fiber development.The physicochemical properties were analyzed using ProtParam tool,phylogenetic trees were constructed with MEGA11.0,chromosomal localization maps were drawn,conserved motifs were analyzed using MEME,promoter cis-acting elements were analyzed using PlantCARE.Transcriptome data was utilized to analyze the tissue expression characteristics of the TUA gene family genes.The results show:there are a total of 67 TUA family members within the four cotton species.The number of these genes in tetraploid cotton species is approximately double that in diploid cotton species,and phylogenetic analysis classified them into five subgroups(A-E).The evolution of the TUA gene family between G.hirsutum and G.barbadense showed similarities.The prediction of cis-acting elements indicates that most of these elements are associated with growth and development,as well as defense against stress.Analysis of the tissue expression characteristics of 23 G.hirsutum TUA genes using TM-1 transcriptome data revealed preferential expression of TUA genes in fibers.Members of the TUA gene family with predominant expression in upland cotton fibers were screened,providing important candidate gene resources for the genetic improvement and breeding of cotton fiber traits.
