摘 要:目的 探讨circ-ANKIB1在心肌肥厚中的作用。方法 (1)构建乳鼠原代心肌细胞肥大模型;(2)检测circ-ANKIB1是线性还是环性;(3)检测circ-ANKIB1的富集情况;(4)检测circ-ANKIB1的表达水平;(5)设计合成si-circ-ANKIB1,检测敲减效率;(6)检测敲减circ-ANKIB1后对心肌肥厚分子标志物的影响。结果 (1)检测肥厚标志物证实心肌肥厚模型构建成功;(2)Sanger测序证实circ-ANKIB1反向剪接成环;(3)RNase R酶处理后富集到约60% circ-ANKIB1;(4)Circ-ANKIB1在心肌细胞肥大模型中的表达有显著改变;(5)在原代心肌细胞中转染针对circ-ANKIB1的siRNA可以敲减70%;(6)敲减circ-ANKIB1后ANP和MYH7的表达水平下调,MYH6的表达水平上调。结论 心肌细胞敲减circ-ANKIB1后ANP和MYH7的表达水平下降,MYH6的表达增加,推测circ-ANKIB1可以促进心肌细胞肥大。Objective To explore the role of circ-ANKIB1 in myocardial hypertrophy.Methods (1) A hypertrophy model of primary cardiomyocytes from neonatal rats was established.(2) The structure of circ-ANKIB1 was identified as either linear or circular.(3) The enrichment of circ-ANKIB1 was assessed.(4) The expression level of circ-ANKIB1 was measured.(5) SiRNAs targeting circ-ANKIB1 were designed and synthesized to evaluate their knockdown efficiency.(6) The impact of circ-ANKIB1 knockdown on molecular markers associated with myocardial hypertrophy was analyzed.Results (1) The myocardial hypertrophy model was successfully established through the detection of hypertrophy markers.(2) Sanger sequencing verified that circ-ANKIB1 is formed through back-splicing into a loop structure.(3) Following RNase R treatment,circ-ANKIB1 was enriched to approximately 60%.(4) In the cardiomyocyte hypertrophy model,the expression of circ-ANKIB1 was significantly altered.(5) Transfection of siRNAs targeting circ-ANKIB1 in primary cardiomyocytes resulted in a reduction of its expression by 70%.(6) Upon circ-ANKIB1 knockdown,the expression levels of ANP and MYH7 were reduced,while the expression of MYH6 was elevated.Conclusion The decrease in ANP and MYH7 expression and the increase in MYH6 expression following circ-ANKIB1 knockdown in cardiomyocytes suggest that circ-ANKIB1 plays a role in promoting cardiomyocyte hypertrophy.
