摘 要:转录后加工是线粒体基因表达调控的重要环节,转录后加工异常将严重影响玉米种子发育.35肽串联重复序列(pentatricopeptide repeat,PPR)蛋白家族等多个蛋白家族参与线粒体基因RNA剪接等转录后加工过程,这些蛋白家族的成员在玉米中尚未全部被鉴定.本研究通过图位克隆鉴定出一个胚乳发育缺陷相关基因ZmEDD1,并对其在玉米种子发育过程中的功能进行了鉴定.ZmEDD1编码一个P型PPR蛋白,命名为玉米胚乳发育缺陷蛋白ZmEDD1.edd1突变体籽粒比B73小,且皱缩,千粒重下降;对授粉后15天(15 days after pollination,15 DAP)和21 DAP的籽粒进行观察发现,edd1突变体籽粒顶部呈现空果壳,胚发育延迟;遗传学分析表明,edd1为单基因隐性突变体,通过图位克隆分析发现,突变基因ZmEDD1定位于1号染色体Bin 1.08.该基因表达产物ZmEDD1蛋白定位于线粒体,且在果穗中表达量最高;ZmEDD1的突变导致nad1等8个线粒体基因的RNA剪接受到影响,同时,edd1线粒体的嵴结构异常.上述研究结果表明,ZmEDD1通过影响线粒体多个基因的RNA剪接参与玉米种子发育.Post transcriptional processing is an important process for regulating mitochondrial gene expression.Abnormal post transcriptional processing will seriously affect the development of maize seeds.Multiple protein families such as pentatricopeptide repeat(PPR)protein family are involved in post-transcriptional processing such as mitochondrial gene RNA splicing.The members of these protein families have not been fully identified in maize.This study identified an endosperm development defect related gene ZmEDD1 through positional cloning and evaluated its function during maize seed development.ZmEDD1 encodes a P-type PPR protein,named maize endosperm developmental defect protein 1(ZmEDD1).The edd1 mutant has smaller seeds than B73,and the seeds are wrinkled with a decreased thousand-grain weight.Observation of the seeds at 15 days after pollination(DAP)and 21 DAP revealed that the edd1 mutant had empty fruit shells at the top of the kernels and delayed embryo development.Genetic analysis indicates that edd1 is a single gene recessive mutant.Mapping analysis revealed that the mutated gene ZmEDD1 was located at Bin 1.08 on chromosome 1,and the ZmEDD1 protein was located in mitochondria and had the highest expression level in the ear.The mutation of ZmEDD1 affected the RNA splicing of 8 mitochondrial genes,including nad1.At the same time,the cristae structure of mitochondria was abnormal in the edd1 mutant.This study indicates that ZmEDD1 may participate in maize seed development by affecting RNA splicing of multiple mitochondrial genes.
