摘  要:为探究绵羊胚胎附植囊胚孵化节点子宫内Th1免疫偏离导致胚胎附植异常的作用机制,在绵羊胚胎附植囊胚孵化节点(第12天)构建正常和内毒素脂多糖(LPS)诱导Th1免疫偏离的妊娠模型,采集母-胎界面子宫内膜组织样本进行转录组测序。通过差异表达基因分析、GO和KEGG富集分析以及蛋白互作网络,筛选关键基因和重要通路。结果表明:Th1免疫偏离的子宫内膜组织,Th1细胞分化细胞因子IL-12和IFN-γ表达显著升高(P<0.05);Th2细胞分化细胞因子IL-4表达显著下降(P<0.05),子宫内膜组织疏松水肿,可见中性粒细胞浸润。共鉴定到424个差异表达基因,如S100A8、S100A9、PTX3和CXCL13等免疫相关基因;差异表达基因显著富集在免疫反应、Toll受体信号、JAK-STAT信号等通路;筛选到20个关键中心基因,1条关键子网,包括经典干扰素刺激基因(ISGs),如ISG15、MX1和MX2等;RT-qPCR结果表明转录组测序数据可靠。The aim of this study was to investigate the mechanism by which Th1 immune deviation in the uterus at the blastocyst hatching node of sheep leads to abnormal embryo implantation.In this study,pregnancy models of normal and LPS-induced Th1 immune deviation at the blastocyst implantation stage(day 12)of sheep embryos were constructed.And Endometrial tissue samples were collected from the maternal-fetal interface for transcriptome sequencing.Key genes and important pathways were identified through differential expression gene analysis,GO and KEGG enrichment analysis,as well as protein interaction network screening.The results showed that within the endometrial tissue of the pregnancy model,there was a shift towards Th1 immune deviation,with a significant upregulation in the expression of Th1 cell differentiation cytokines IL-12 and IFN-γ(P<0.05),and a significant downregulation in the expression of Th2 cell differentiation cytokine IL-4(P<0.05).Additionally,the endometrial tissue exhibited looseness and edema,accompanied by neutrophil infiltration.A total of 424 differentially expressed genes were identified,such as immune-related genes S100A8,S100A9,PTX3,and CXCL13.The differentially expressed genes were significantly enriched in pathways such as immune response,Toll-like receptor signaling pathway,and JAK-STAT signaling pathway.Twenty key central genes and one key subnet were identified,including classical interferonstimulated genes(ISGs),such as ISG15,MX1,and MX2.The RT-qPCR results confirmed the reliability of the transcriptome sequencing data.
