摘 要:骤然降温和极端低温等低温胁迫影响紫花苜蓿(Medicago sativa L.)的种植和生产,严重影响着我国饲草产业的健康发展。本研究选择‘新疆大叶’和‘肇东’两种秋眠性不同的紫花苜蓿进行24 h和192 h时长的低温处理,通过转录组测序技术筛选冷响应基因。经过192 h的低温处理,秋眠型的‘肇东’在192 h时抗寒性显著高于半秋眠型‘新疆大叶’。通过0 h和24 h两个时间点的转录组测序分析,进一步筛选出13个与抗寒性相关的差异表达基因。经过qRT-PCR荧光定量验证后,有7个基因的表达趋势与转录组数据一致,分别为两品种紫花苜蓿中表达趋势相反的2个基因和在两品种紫花苜蓿中表达趋势相同的5个基因。此外,对所有差异表达基因进行京都基因和基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路富集分析,找出淀粉和蔗糖代谢、撕裂原活化蛋白激酶(Mitogen-Activated Protein Kinase,MAPK)信号途径和半乳糖代谢等11条与抗寒相关的代谢通路。本研究筛选到的差异基因和代谢通路,将为后续紫花苜蓿抗寒机制及抗寒分子育种的研究提供参考。The cultivation and production of Medicago sativa L.are affected by abrupt cold and extreme low temperature,which seriously affect the healthy development of forage industry in China.In this study,two cultivars of alfalfa with different fall dormancy,‘Xinjiang Daye’and‘Zhaodong’,were treated with low temperature for 24 h and 192 h,and cold response genes were screened by transcriptome sequencing technology.After 192 h of low temperature treatment,the cold resistance of‘Zhaodong’was significantly higher than that of‘Xinjiang Daye’at 192 h.Through transcriptome sequencing analysis at 0 h and 24 h,13 differentially expressed genes related to cold resistance were further screened.After qRT-PCR fluorescence quantification was verified,7 genes with expression patterns consistent with the transcriptome data were identified:2 genes with opposite expression trends in the two varieties of alfalfa,and 5 genes with the same expression trend in the two varieties of alfalfa.In addition,KEGG pathway enrichment analysis was performed on all differentially expressed genes,and 11 metabolic pathways related to cold resistance including starch and sucrose metabolism,MAPK signaling pathway and galactose metabolism were identified.The differential genes and metabolic pathways screened in this study would provide references for subsequent studies on cold resistance mechanism and molecular breeding of alfalfa.
