抗原减弱与不规则抗体造成ABO血型正反不一致原因分析

摘　　要：目的 分析两例ABO血型异常的原因,得到正确的血型结果。方法 应用常规血清学方法检测ABO血型,并采用分子生物学方法对其外显子DNA序列进行测定。同时对两例样本的血浆进行红细胞不规则抗体筛选。结果 两例血型均为亚型,其中1例为Ael05,血浆中含有抗-A1抗体;另1例为Bel09,血浆中含有抗-M抗体。结论 ABO血型检测中,应正确使用O型细胞,必要时应用基因分型技术,准确定型对输血安全性至关重要。Objective To analyze the reasons of ABO discrepancies in two cases and get the correct blood group results.Methods ABO blood group was detected by routine serological method,and exon DNA sequence was determined by direct sequencing method.The irregular antibodies screening was carried out for 2 samples.Results Both blood groups samples were ABO subgroup,sample 1 was Ael05,and the plasma contained anti-A1 antibody;sample 2 was Bel09,and the plasma contained anti-M antibody.Conclusion In ABO blood type testing,O-type cells should be used correctly.When necessary,genotyping techniques should be applied.Accurate typing is crucial for the safety of blood transfusion.
