基于Wnt通路探讨乌司他丁对膀胱癌细胞上皮间质转化的影响

摘　　要：目的探究乌司他丁对膀胱癌T24细胞上皮间质转化的影响。方法将膀胱癌T24细胞随机分为空白组、对照组、实验组和联合组。空白组给予正常培养,对照组给予含0.75μmol·L^(-1)KYA1797K的RPMI-1640培养基进行处理;实验组给予含5.00μmol·L^(-1)乌司他丁的RPMI-1640培养基进行处理,联合组给予含5.00μmol·L^(-1)乌司他丁与0.75μmol·L^(-1)KYA1797K的RPMI-1640培养基进行处理。干预48 h后,用四甲基偶氮唑蓝法检测细胞的存活率,用Transwell实验法检测细胞的侵袭与迁移能力,用蛋白质印迹法检测上皮间质转化相关蛋白、整合位点家族成员3α(Wnt3α)和β连环蛋白(β-catenin)的表达水平。结果空白组、对照组、实验组和联合组的细胞存活率分别为(100.00±0.00)%、(73.28±6.37)%、(76.98±5.82)%和(40.36±3.21)%,侵袭细胞数量分别为(66.83±8.18)、(33.33±3.27)、(38.67±4.32)和(11.33±1.21)个,细胞迁移数分别为(79.33±6.38)、(53.00±6.26)、(52.33±3.14)和(28.33±5.28)个,E-钙黏蛋白相对表达水平分别为0.23±0.08、0.51±0.02、0.58±0.05和0.82±0.03,波形蛋白相对表达水平分别为0.78±0.03、0.55±0.02、0.58±0.06和0.27±0.06,N-钙黏蛋白相对表达水平分别为0.83±0.08、0.59±0.02、0.62±0.02和0.31±0.09,Wnt3α蛋白相对表达水平分别为0.85±0.09、0.28±0.13、0.29±0.05和0.12±0.01,β-catenin蛋白相对表达水平分别为0.81±0.05、0.25±0.07、0.22±0.03和0.08±0.02。实验组、对照组的上述指标分别与空白组、联合组相比较,在统计学上差异均有统计学意义(均P<0.05)。结论乌司他丁能够抑制膀胱癌T24细胞的上皮间质转化过程,其作用机制可能与抑制Wnt信号通路相关。Objective To investigate the effect of ulinastatin on epithelial-mesenchymal transition(EMT) of bladder cancer T24 cells. Methods Bladder cancer T24 cells were randomly divided into blank, control, experimental, combined groups. The cells in the blank group were cultured normally;and the cells in the control group were treated with RPMI-1640 medium containing 0.75 μmol·L^(-1) KYA1797K;the cells in the experimental group were treated with RPMI-1640 medium containing 5.00 μmol·L^(-1) ulinastatin and the cells in the combined group were treated with RPMI-1640 medium containing 5.00 μmol·L^(-1) ulinastatin and 0.75 μmol·L^(-1) KYA1797K. The treatment time was 48 hours. The cell viability was detected by methyl thiazolyl tetrazolium (MTT) assay. Transwell assay was used to detect the invasion and migration ability of cells. The protein expression levels of epithelial-mesenchymal transition related proteins,wingless-type MMTV integration site family 3α( Wnt3α)and β-catenin were detected by Western blotting.
