摘 要:试验旨在探究复合脂肪酸对骡鸭原代肝细胞超微结构、脂质沉积、氧化应激及AMPK信号通路相关基因表达的影响。试验使用不同浓度的复合酸(油酸+棕榈酸)进行处理:500μmol/L+250μmol/L、1000μmol/L+500μmol/L、1500μmol/L+750μmol/L和2000μmol/L+1000μmol/L,并持续处理36 h构建原代骡鸭肝细胞脂质沉积模型。结果显示:(1)分离的骡鸭原代肝细胞有丰富糖原颗粒和脂滴的存在。(2)与对照组相比,油酸+棕榈酸浓度为1500μmol/L+750μmol/L时,培养骡鸭原代肝细胞36 h,细胞活力没有显著差异(P>0.05),脂滴数量显著增加(P<0.05)。(3)透射电镜结果显示,复合脂肪酸处理的骡鸭原代肝细胞和线粒体均出现明显水肿,脂滴出现聚合,并且膜和嵴的结构受损。此外,与对照组相比,肝细胞脂滴含量、谷丙转氨酶和谷草转氨酶活性、丙二醛和活性氧水平显著上升(P<0.05)。(4)与对照组相比,复合脂肪酸处理的骡鸭原代肝细胞中AMP活化蛋白激酶(AMPK)、脂蛋白脂肪酶(LPL)和肉碱棕榈转移酶-1A(CPT-1A)mRNA表达量以及AMPK蛋白表达量显著降低(P<0.05),而白细胞分化抗原36(CD36)、乙酰辅酶A羧化酶1(ACC1)mRNA及白细胞分化抗原36(CD36)、胆固醇调节元件结合蛋白1(SREBP1)蛋白表达量显著提高(P<0.05)。因此,油酸+棕榈酸浓度为1500μmol/L+750μmol/L处理骡鸭原代肝细胞36 h能通过调控AMPK信号通路,有效增加肝细胞脂质沉积,但同时易诱导肝细胞氧化应激。This experiment aimed to explore the effects of compound fatty acids on fat deposition,ultrastructure,oxidative stress and the expression of genes related to AMPK signaling pathway in mule duck primary hepatocytes.By constructing a model of lipid deposition of mule duck primary hepatocytes,different concentrations of mixed acids(oleic acid+palmitic acid),the concentrations were 500μmol/L+250μmol/L,1000μmol/L+500μmol/L,1500μmol/L+750μmol/L,2000μmol/L+1000μmol/L,the lipid deposition model of primary mule duck hepatocytes was constructed after 36 hours of continuous treatment.The results showed that:①Isolated mule duck primary hepatocytes had the presence of abundant glycogen particles and lipid droplets.②When the ratio of oleic acid to palmitic acid reached 1500μmol/L+750μmol/L,there was no significant difference in cell viability(P>0.05),Oil Red O staining revealed a significant increase in lipid droplets content in hepatocytes(P<0.05).③Transmission electron microscopy revealed that significant edema appeared in both cells and mitochondria of the hepatocyte lipid deposition model,with aggregation of lipid droplets and damage to the structure of membranes and cristae.And there were significant increases in the activities of glutamic pyruvic transaminase and glutamic oxaloacetic transaminase,malondialdehyde and reactive oxygen level compared with the control group(P<0.05).④Compared with the control group,the mRNA expression levels of AMP-activated protein kinase(AMPK),lipoprotein lipase(LPL)and carnitine palmitoyl transferase-1A(CPT-1A)and the AMPK protein expression level in mule duck primary hepatocytes treated with composite fatty acids were significantly lower(P<0.05),while the mRNA expression levels of leukocyte differentiation antigen 36(CD36),acetyl CoA carboxylase 1(ACC1)and the protein expression levels of leukocyte differentiation antigen 36(CD36)and sterol-regulatory element binding protein 1(SREBP1)were significantly higher(P<0.05).In summary,treatment of mule duck primary hepatocytes w
