摘 要:研究旨在通过比较豁眼鹅正常眼和豁眼个体的胚胎期眼睑转录组,筛选豁眼性状的关键候选基因。试验选择18胚龄的豁眼鹅胚胎,分别挑选出3只豁眼和3只正常眼个体,采集发生缺损部位眼睑组织,进行转录组测序和分析。结果显示:豁眼组与正常眼组相比,共有252个差异表达基因,其中上调基因有92个,下调基因160个;结合GO和KEGG数据库对DEGs进行功能注释和通路分析显示,DEGs显著富集到细胞迁移、胞葬作用、神经活性配体—受体相互作用、免疫系统过程等信号通路;进一步结合PPI网络分析等手段,筛选出TACR2、CCL19、DIO1、LRIT3等可能与鹅豁眼性状相关的候选基因。研究表明,豁眼鹅眼睑缺损性状的形成可能与细胞迁移、胞葬等信号通路有关,相关通路中的TACR2、CCL19、DIO1、LRIT3基因可能是眼睑缺损性状的关键基因。The study was aimed to compare the transcriptome of eyelid tissue from Huoyan goose with congenital eyelid colobomas(CEC)or normal eyelids during the embryonic period and to identify the key genes associated with CEC.To collect eyelid tissues for transcriptome,three individuals with CEC eyelid or norm eyelid were selected from embryos of Huoyan geese at 18 d.The results showed that compared with the geese with normal eyelid,there were 252 differentially expressed genes,including 92 up-regulated genes and 160 down-regulated genes.Functional annotation and KEGG pathway analysis of DEGs showed that DEGs were mainly enriched in pathways,such as cell motility,efferocytosis,neuroactive ligand-receptor interaction and immune system processes,etc.By protein-protein interaction(PPI)network analysis,genes such as TACR2,CCL19,DIO1 and LRIT3,were identified as candidate genes.The results suggested that the formation of CEC in Huoyan goose might be related with the the pathways such as cell motility,efferocytosis,and the genes such as TACR2,CCL19,DIO1,LRIT3 that involved in those pathways might be the key gene of CEC formation.
