摘 要:目的:对金银花药材各化学成分进行数据分析,建立谱-效相关金银花药材抑菌作用质量评价体系。方法:通过考察流动相、波长、流速等条件,确定最佳液相色谱条件,建立50批金银花药材的定量指纹图谱;通过微量稀释法得出金银花药材的最佳抑菌浓度,并对50批样品进行抑菌测定,得出抑菌率;采用相似度分析、灰色关联度分析及通过支持向量机法建立数学模型等对50批定量指纹图谱及抑菌率结果进行分析。结果:建立的50批金银花药材定量指纹图谱中选择了21个共有峰,相似度在0.343~1,确立了6个化学成分(峰4为新绿原酸、峰9为绿原酸、峰10为隐绿原酸、峰18为异绿原酸B、峰20为异绿原酸A、峰21为异绿原酸C),50批金银花药材的抑菌率平均值在0.310 3%~98.64%。灰色关联度分析显示仅成分1抑菌作用呈负相关,其余均呈正相关。数学模型的拟合数据与实验数据的相对偏差均在4%以下。结论:建立的金银花药材液相色谱条件稳定可靠,建立的金银花药材抑菌作用质量评价体系能够达到从抑菌率评价药物质量的目的。OBJECTIVE To establish a quality evaluation system for the antibacterial activity of Flos Lonicerae by analyzing the data of chemical components.METHODS By studying the mobile phase,wavelength,flow rate and other conditions,the optimal liquid chromatography conditions were determined,and the quantitative fingerprint of 50 batches of Flos Lonicerae was established.The optimal antibacterial concentration of Flos Lonicerae was obtained by a microdilution method,and the antibacterial rate of 50 batches of samples was determined.The results of 50 batches of quantitative fingerprints and antibacterial rate were analyzed by the similarity analysis,grey correlation analysis and mathematical modeling by support vector machine.RESULTS Twenty-one common peaks were selected from the 50 batch quantitative fingerprints of Flos Lonicerae,and the similarity was between 0.343 to 1.Six chemical components were identified(peak 4:neochlorogenic acid,peak 9:chlorogenic acid,peak 10:cryptochlorogenic acid,peak 18:isochlorogenic acid B,peak 20:isochlorogenic acid A,peak 21:isochlorogenic acid C).The average antibacterial rate of 50 batches of Flos Lonicerae ranged from 0.3103%to 98.64%.The results of grey correlation analysis showed that only component 1 was negatively correlated with antibacterial activity,and the rest were positively correlated.The relative deviation between the fitting data of the mathematical model and the experimental data was less than 4%.CONCLUSION The established liquid chromatography conditions of Flos Lonicerae are stable and reliable,and the established antibacterial quality evaluation system of Flos Lonicerae can achieve the purpose of evaluating the quality of drugs from the antibacterial rate.
