摘 要:为了探究自噬在松材线虫(Bursaphelenchus xylophilus)抵御松树分泌的蒎烯胁迫过程中的作用,用不同质量浓度的α-蒎烯和β-蒎烯混合液体外处理线虫,模拟树体胁迫,观察蒎烯胁迫对线虫取食和繁殖的影响,克隆松材线虫自噬基因BxATG6、BxATG10和BxATG12,检测胁迫后这3个基因的表达情况,分析松材线虫自噬基因在抵御蒎烯胁迫中的作用。结果显示:松材线虫自噬基因BxATG6、BxATG10和BxATG12编码的蛋白与稻叶牙滑刃线虫(Aphelenchoides besseyi)相关自噬蛋白的亲缘关系较近。低质量浓度蒎烯混合液(32.1 g·L^(-1))会显著抑制线虫的取食和繁殖,胁迫3 h以上,线虫3个自噬基因的表达量显著上调。由此表明,松材线虫自噬基因BxATG6、BxATG10和BxATG12在抵御蒎烯胁迫中起着一定的作用。In order to explore the role of autophagy in Bursaphelenchus xylophilus during its resistance to the pinene stress secreted by pine trees,nematodes were treated in vitro with mixed solutions ofα-pinene andβ-pinene at different mass concentrations to simulate the stress in trees.The effects of pinene stress on the feeding and reproduction of nematodes were observed.The autophagy genes BxATG6,BxATG10,and BxATG12 of B.xylophilus were cloned,and the expression levels of these three genes after stress were detected,so as to analyze the role of the autophagy genes of B.xylophilus in resisting pinene stress.The results showed that the proteins encoded by the autophagy genes BxATG6,BxATG10,and BxATG12 of B.xylophilus had a relatively close genetic relationship with the related autophagy proteins of Aphelenchoides besseyi.A low mass concentration of the pinene mixed solution(32.1 g·L^(-1))significantly inhibited the feeding and reproduction of nematodes.After being stressed for more than 3 hours,the expression levels of the three autophagy genes of nematodes were significantly up-regulated.This indicates that the autophagy genes BxATG6,BxATG10,and BxATG12 of B.xylophilus play a certain role in resisting pinene stress.
