摘 要:【目的】细菌性溃疡病是全球猕猴桃种植区共同面临的严重病害。筛选抗性种质资源、提高品种抗病性是控制猕猴桃溃疡病蔓延的重要手段。‘金奉’猕猴桃是课题组从本地选育的优良品种,评价其实生后代群体对溃疡病的抗性,可为培育抗性种质、改良品种提供材料基础。【方法】以171份‘金奉’猕猴桃实生后代为材料,运用离体枝条和离体叶盘人工接种溃疡病菌方法对其抗性进行初步评价与进一步验证,并且对其中抗性差异显著的个体进行抗性相关生理生化指标的测定。【结果】相关性分析表明,枝条接种法与叶盘法结果呈正相关。基于枝条病斑长度和叶盘病斑占比,171份猕猴桃实生后代种质资源可划分为抗病、耐病、感病及高感4个级别,其中抗病种质49份,耐病种质62份,感病种质49份,高感种质11份。从中选出11份抗性差异显著的种质,进一步进行生理生化指标测定,发现其叶片生理指标中的木质素、可溶性糖和总酚含量与个体抗性呈显著正相关。【结论】离体枝条、叶盘法2种抗性评价结果显著相关,叶片生理指标测定与抗性鉴定结果显著相关,证实了离体枝条和叶盘人工接种法是可靠的,3种叶片的生理指标测定可用于抗性种质的快速鉴定。筛选到的抗病种质为‘金奉’猕猴桃的溃疡病抗性育种奠定了材料基础。[Objective]Bacterial canker is a serious disease of kiwifruit around the world.Screening resistant germplasm resources and improving disease resistance of kiwifruit are important means to control the spread of bacterial canker disease.‘Jinfeng’kiwifruit is an excellent local cultivar bred by our research group.The resistance to bacterial canker disease of‘Jinfeng’kiwifruit seed offspring was evaluated and this can lay the material foundation for selecting resistant germplasm and improving varieties.[Method]The 171 seed offspring of‘Jinfeng’kiwifruit were used as materials.The resistance was evaluated and further verified by the methods of artificial inoculation of canker bacteria in isolated branches and leaf disc in vitro.11 seed offspring with significant resistance differences were determined resistance-related physiological and biochemical indices.[Result]Correlation analysis showed that the branch inoculation method was positively correlated with the leaf disc method.Based on the length of branch disease spots and the proportion of leaf disc disease spots,171 samples can be divided into four levels:disease resistance,disease tolerance,disease sensitivity and high sensitivity,among which 49 samples were disease resistance resources,62 samples were disease tolerance resources,49 samples were disease sensitivity resources and 11 samples were disease high sensitivity resources.From them,11 seed offspring with resistance differences were selected and further determined by physiological and biochemical indicators,and found that the total phenol,soluble sugar and lignin content in the leaf physiological indexes showed significantly positive correlations with the individual resistance.[Conclusion]The results of resistance evaluation in isolated branches and leaf disc method were significantly correlated,and the determination of leaf physiological indicators were significantly correlated with the results of resistance identification,which confirmed the manual inoculation method of isolated branches and is
