摘  要:绵羊感染巨型住肉孢子虫后会出现呕吐和肠炎等症状,给畜牧业带来巨大经济损失。以巨型住肉孢子虫全蛋白为抗原,优化各项反应条件,建立一种耗时短、成本低且可在动物生前进行操作的巨型住肉孢子虫的间接ELISA检测方法。用建立的ELISA方法对青海省6个地区538份藏羊血清样品进行巨型住肉孢子虫的检测。结果显示,当包被浓度0.05μg、血清稀释比例为1∶100、二抗稀释比例为1∶4000时重复性良好。青海省6个地区藏羊血清IgG和IgM阳性率分别为16.2%和20.3%。结果表明,利用巨型住肉孢子虫的全蛋白成功建立了间接ELISA检测方法,该方法具有良好的重复性,可用于临床对藏羊巨型住肉孢子虫感染的初步检测。Sheep infected with Sarcocystis gigantea will have symptoms such as vomiting and enteritis,which will bring huge economic losses to the livestock industry.In this study,the whole protein of Sarcocystis gigantea was used as the antigen to optimize the reaction conditions,and an indirect ELISA method for the detection of Sarcocystis gigantea was established with short time,low cost and can be operated before the animal's death.The established ELISA method was used to detect Sarcocystis gigantea in 538 Tibetan sheep serum samples from 6 regions of Qinghai province.The results showed that the coating concentration was 0.05μg,the serum dilution ratio was 1∶100,and the secondary antibody dilution ratio was 1∶4000.The positive rates of serum IgG and IgM in Tibetan sheep in six regions of Qinghai province were 16.2%and 20.3%,respectively.The results showed that an indirect ELISA method was successfully established using the whole protein of Sarcocystis gigantea,which had good reproducibility and could be used for the preliminary detection of Sarcocystis gigantea infection in Tibetan sheep.
