摘  要:孔蛋白在免疫原和毒力决定因素方面有重要作用。为探究沙门氏菌中孔蛋白ompD、ompS、ompW基因突变是否对沙门氏菌的致病性产生影响,在构建鼠伤寒沙门氏菌孔蛋白突变株STM LT2ΔompD、STM LT2ΔompS、STM LT2ΔompW的基础上,测定了突变株对其小鼠致病性、体内遗传稳定性、LD 50、肝脏和脾脏载菌量。结果表明,标准菌株和突变株均对小鼠产生致病性,肝脏和脾脏体积明显增大;突变株在小鼠体内连续传代20次仍然能够稳定遗传;LD 50毒力测定发现突变株的毒力均呈现减弱趋势,其中突变株STM LT2ΔompS毒力减弱最为明显;肝脾载菌量测定发现,相比标准菌株,突变株在小鼠肝、脾上分布的细菌含量均明显减少。试验结果为鼠伤寒沙门氏菌减毒疫苗开发奠定基础,也可为探讨鼠伤寒沙门氏菌致病作用的分子机制提供参考。Pore proteins have important roles in immunogenicity and virulence determinants.In order to investigate whether mutations in the pore protein ompD,ompS,and ompW genes in Salmonella have an effect on the pathogenicity of Salmonella,this study was conducted on the basis of constructing Salmonella typhimurium pore protein mutant strains STM LT2ΔompD,STM LT2ΔompS,and STM LT2ΔompW,and its mouse pathogenicity,in vivo genetic stability,LD 50,and hepatic and splenic loads were were determined.The results showed that both the standard strain and the mutant strains were pathogenic to mice,and the sizes of liver and spleen increased significantly;the mutant strains were still genetically stable in mice for 20 consecutive generations;the LD 50 toxicity assay revealed that the virulence of the mutant strains showed a trend of weakening,with the most obvious weakening of the virulence of the mutant strain STM LT2ΔompW;and the liver and spleen load assay revealed that,compared with the standard strain,the mutant strains were distributed in the liver and spleen of mice,with a greater distribution in the liver and spleen.The bacterial loads in the liver and spleen of the mutant strains were found to be significantly reduced compared with the standard strain.This experiment lays the foundation for the development of Salmonella typhimurium attenuated vaccine,and also provides a reference for exploring the molecular mechanism of Salmonella typhimurium pathogenicity.
