鸽源金黄色葡萄球菌外毒素基因与细菌分子分型相关性研究

摘  要:旨在对鸽源金黄色葡萄球菌(Staphylococcus aureus)的毒素基因与agr、spa基因型进行研究,并分析其分型与毒素基因之间的相关性。利用多重PCR检测分离菌株的辅助基因调节子并对其进行蛋白A测序。检测表明,毒素基因中hla、hlb基因检出率较高;其次为sec、seg、seh检出率均为16.00%;sea、seb、sed、see、tst检出率最低,均为2.00%;MRSA菌株携带4种及以上毒素基因。分型显示,S.aureus的agr分型中,agrⅠ型为鸽源S.aureus优势菌株,占分离率的70.00%;agrⅡ和agrⅢ检出率分别为12.00%、18.00%;agrⅣ未检出。spa共有9种分型,t011 S.aureus是鸽源主要的流行菌株,占68.00%,其次为t127、t502均为10.00%。agrⅡ和agrⅢS.aureus更具有携带多种毒素基因的潜力,而agrⅠ型毒素基因携带能力较弱;t011型S.aureus均携带hla基因,t502和t1710中的MRSA菌株均携带7种毒素基因;说明同种分型携带毒素基因情况具有一致性。t011均携带agrⅠ,t127均携带agrⅢ,t502均携带agrⅢ,说明spa与agr有一定相关性。研究通过S.aureus分型检测,分析其潜在携带毒素基因的能力,为鸽源S.aureus感染性疾病的监控及分子流行病学的研究提供科学依据。To study the toxin genes,agr and spa genotypes of Staphylococcu aureus derived from pigeons and to analyze the correlation between its typing and toxin genes,the agr typing of the isolates was detected by multiplex PCR,and spa sequencing was performed.The results showed that:(1)The detection rate of hla and hlb in toxin genes were high;The detection rates of sec,seg and seh were 16.00%.The lowest detectable rates of sea,seb,sed,see and tst were 2.00%;MRSA strains carry four or more toxin genes.(2)Genotyping results showed that among the agr subtypes of S.aureus,agrⅠwas the dominant strain of S.aureus derived from pigeons,accounting for 70.00%of the isolation rate;The detection rates of agrⅡand agrⅢwere 12.00%and 18.00%,respectively.agrⅣwas not detected.Nine subtypes were detected by spa gene.t011 type S.aureus was the main epidemic strain of pigeon origin,accounting for 68.00%,followed by t127 and t502,which were both 10.00%.agrⅡand agrⅢS.aureus have more potential to carry multiple toxin genes,while the carrying capacity of agrⅠtoxin gene is weak.S.aureus of type t011 all carry the hla gene,and the MRSA strains of types t502 and t1710 all carry seven toxin genes,indicating that condition of the toxin genes carried by the isotypes is consistent.t011 all carried agrⅠ,t127 and t502 all carried agrⅢ.It indicated that there was a correlation between spa classification and agr classification.In this study,the potential ability to carry toxin genes was analyzed by S.aureus typing test,which would provide a scientific basis for the monitoring of pigeon-derived S.aureus infections and the research of molecular epidemiology.
