摘 要:目的研究银莲花总皂苷(TSP)对人肝癌HepG2细胞增殖的抑制作用及机制。方法体外培养人肝癌HepG2细胞,将细胞分为空白对照(Control)组和TSP组(6.25、12.5、25、50、100、200μg/mL)。CCK-8试验检测细胞增殖抑制率;流式细胞术检测细胞周期和凋亡率;Western blot法检测Bcl-2、Bax和Caspase-3蛋白表达;试剂盒检测HepG2细胞内谷胱甘肽过氧化物酶(GSH-px)、超氧化物歧化酶(SOD)、活性氧(ROS)水平及丙二醛(MDA)表达。结果与Control组比较,TSP在12.5~200μg/mL浓度范围内对HepG2细胞增殖均有明显的抑制作用(P<0.05,P<0.01),且呈现浓度依赖性;TSP 25、50、100μg/mL组HepG2细胞凋亡率均明显上升(P<0.01),G0/G1期细胞比例均明显增加(P<0.05,P<0.01),S期比例均明显降低(P<0.05,P<0.01);TSP 25、50、100μg/mL组均能明显下调Bcl-2蛋白表达(P<0.01),上调Bax、Caspase-3蛋白表达(P<0.01);TSP 50、100μg/mL组能明显降低HepG2细胞内GSH-px、SOD水平(P<0.05,P<0.01),增加ROS、MDA水平(P<0.01)。结论TSP能浓度依赖性抑制HepG2细胞增殖,将细胞周期阻滞于G0/G1期,并诱导细胞凋亡,作用机制可能与其能促进细胞产生氧化损伤和调控细胞凋亡相关蛋白表达有关。Objective To study the effect of total saponins of pasqueflower(TSP)on the proliferation of HepG2 cells and its mechanism.Methods HepG2 cells were cultured in vitro.The cells were divided into control group and TSP 6.25,12.5,25,50,100,200μg/mL groups.CCK-8 assay was used to detect the effect of TSP on HepG2 cell proliferation.The effects of TSP on cell cycle and apoptosis were detected by flow cytometry.Western blot was used to detect Bcl-2,Bax and Caspase-3 protein expression.GSH-px,SOD,ROS and MDA expression in HepG2 cells were detected by the kits.Results Compared with the control group,the proliferation inhibition rate of HepG2 cells was significantly increased by TSP at 12.5-200μg/mL(P<0.05,P<0.01),which was concentration-dependent.The apoptosis rates of HepG2 cells in TSP 25,50 and 100μg/mL groups were significantly increased(P<0.01),the proportion of G0/G1 phase cells was significantly increased(P<0.05,P<0.01),and the proportion of S phase cells was significantly decreased(P<0.05,P<0.01).The protein expression levels of Bax,Caspase-3 were up-regulated significantly in TSP 25,50,100μg/mL groups(P<0.01);while the expression level of Bcl-2 was down-regulated significantly(P<0.01).TSP 50 and 100μg/mL groups significantly decreased the levels of GSH-px and SOD(P<0.05,P<0.01),and increased the levels of ROS and MDA in HepG2 cells(P<0.01).Conclusion TSP can inhibit HepG2 cell proliferation and induce apoptosis in a concentration-dependent manner,and block the cell cycle in G0/G1 phase.The mechanism of the above effects may be related to the promotion of oxidative damage and the regulation of apoptosis-related protein expression.
