摘 要:目的基于代谢组学探讨肾茶对高尿酸血症(HUA)小鼠的保护作用及相关机制。方法全自动生化分析仪检测小鼠血清尿酸(UA)、肌酐(Cr)和尿素氮(BUN)水平;HE观察肾组织病理变化情况;ELISA试剂盒检测肝脏黄嘌呤氧化酶(XOD)的含量;非靶向代谢组学技术分析HUA小鼠血清代谢谱,结合多元统计学方法筛选差异代谢物,通过KEGG富集分析获得差异代谢通路。结果与空白组比较,模型组小鼠血清中UA、Cr、BUN含量及肝脏组织中XOD含量均显著升高,模型组小鼠肾脏组织可见肾小球皱缩,肾小管管腔扩大;与模型组比较,肾茶组小鼠血清UA、Cr、BUN含量及肝脏组织中XOD含量均有所降低,肾茶及阳性药非布司他对小鼠肾脏组织损伤均有不同程度改善;非靶向代谢组学结果显示,给予肾茶后,血清中16种差异代谢物水平均有不同程度的回调,主要涉及色氨酸代谢、甘油磷脂代谢、嘧啶代谢通路。结论肾茶对HUA具有一定改善作用,其机制可能与色氨酸代谢、甘油磷脂代谢、嘧啶代谢等生物途径相关。Objective To investigate the protective effect of Orthosiphon aristatus on hyperuricemia(HUA)mice and related mechanisms based on metabolomics.Methods Fully automatic biochemical analyser was used to detect serum uric acid(UA),creatinine(Cr)and urea nitrogen(BUN)levels in mice;HE was used to observe pathological changes in renal tissues;ELISA kit was used to detect the content of XOD in liver;non-targeted metabolomics technology was used to analyze serum metabolic profiles of HUA mice,combined with multivariate statistics to screen for different metabolites,and differential metabolic pathways were obtained by KEGG enrichment analysis.Results Compared with the blank group,the serum levels of UA,Cr,BUN and the XOD content in liver tissue of mice in the model group were significantly elevated,and the renal tissue of mice in the model group could be seen to have crumpled glomeruli and enlarged tubular lumen of renal tubules;compared with the model group,the serum levels of UA,Cr,BUN and the XOD content in liver tissue of mice in the Orthosiphon aristatus group were reduced,and Orthosiphon aristatus and the positive drug Febuxostat ameliorated the damage of kidney tissue in different degrees;the results of non-targeted metabolomics showed that the levels of 16 differential metabolites in serum were differentially regulated after administration of renal tea,mainly involving tryptophan metabolism,glycerophospholipid metabolism,and pyrimidine metabolism pathway.Conclusion Orthosiphon aristatus has an ameliorative effect on HUA,and the mechanism may be related to tryptophan metabolism,glycerophospholipid metabolism,pyrimidine metabolism,and other biological pathways.
