摘 要:目的探讨锦灯笼提取物片剂对急性咽炎大鼠的作用及机制。方法将大鼠随机分为空白组、模型组、阳性组、锦灯笼提取物片剂低、中、高剂量组,每组8只。采用向大鼠咽喉喷15%氨水的方法建立急性咽炎大鼠模型。各组大鼠灌胃给药,每日2次,持续7 d。造模及给药期间每天观察并记录大鼠一般情况,并对咽喉组织情况进行评分;采用苏木素-伊红(HE)染色法观察大鼠咽部组织病理学变化;ELISA法检测大鼠血清白细胞介素6(IL-6)、白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)和前列腺素E2(PGE2)含量;血常规分析仪检测白细胞、淋巴细胞、单核细胞以及中性粒细胞数;Western blot法检测大鼠咽部组织中环氧合酶-2(COX-2)、核转录因子κB(NF-κB)、磷酸化NF-κB(p-NF-κB)、核转录因子κB抑制蛋白(Iκ-B)及p-Iκ-B表达水平。结果与空白组比较,模型组大鼠咽部红肿程度显著增加,咽部组织评分显著升高(P<0.05);血清炎症因子IL-1β、IL-6、TNF-α和PGE2水平均显著上升(P<0.05);白细胞、单核细胞以及中性粒细胞数显著增加(P<0.05),淋巴细胞数也增加,但差异无统计学意义(P>0.05)。与模型组比较,阳性组、锦灯笼给药高剂量、中剂量组大鼠的咽喉组织评分显著下降(P<0.05),阳性组和高剂量组口腔分泌量显著下降(P<0.05),阳性组和高剂量组血清炎症因子IL-1β、IL-6、TNF-α和PGE2水平均显著下降(P<0.05),同时血常规分析得出阳性组、高剂量组和中剂量组白细胞、中性粒细胞数显著降低(P<0.05),低剂量组的淋巴细胞数、单核细胞数和中性粒细胞数显著降低(P<0.05),Western blot检测出锦灯笼组的NF-κB、p-NF-κB、p-Iκ-B、COX-2蛋白表达水平均显著下调(P<0.05),Iκ-B蛋白显著上调(P<0.05)。结论锦灯笼提取物片剂可显著改善急性咽炎大鼠的症状,其机制可能与调控NF-κB/COX-2信号通路有关。Objective To explore the effect and mechanism of the tablet of Physalis alkekengi L.extract on rats with acute pharyngitis.Methods A rat model of acute pharyngitis was established by spraying 15%ammonia into the throat of rats.The rats were randomly divided into blank group,model group,and low,medium and high dose group,with 8 rats in each group.Rats in each group were given the drug by gavage twice daily for 7 d.The general conditions of the rats were observed and recorded every day during the modeling and drug administration period,and the pharyngeal tissues were scored,histopathological changes in the pharynx of the rats were observed by hematoxylin-eosin(HE)staining,serum levels of interleukin 6(IL-6),interleukin 1 beta(IL-1β),tumor necrosis factorα(TNF-α)and prostaglandin E2(PGE2)in the rats were detected by ELISA.Leukocytes,lymphocytes,monocytes and neutrophils were measured by blood analyzer,and Western blot was used to detect the expression levels of cyclooxygenase-2(COX-2),nuclear factor-κB(NF-κB),p-NF-κB,inhibitor of NF-κB(Iκ-B)and p-Iκ-B in pharyngeal tissue.Results Compared with the blank group,the pharynx of rats in the model group was significantly more red and swollen,and the pharyngeal tissue score was significantly higher(P<0.05),the levels of serum inflammatory factors IL-1β,IL-6,TNF-αand PGE2 were significantly increased(P<0.05),the numbers of leukocytes,monocytes and neutrophils were significantly increased(P<0.05),and the numbers of lymphocytes were alsoincreased,but the differences were not statistically significant(P>0.05).Compared with the model group,the pharyngeal tissue scores of the rats in the positive group,the high dose and the medium dose groups decreased significantly(P<0.05),the oral secretion of the rats in the positive group and the high dose group decreased significantly(P<0.05),and the levels of the serum inflammatory factors IL-1β,IL-6,TNF-αand PGE2 decreased significantly in the positive and the high dose groups(P<0.05),and at the same time,the blood analysis s
