摘  要:为建立小黑桫椤(Gymnosphaera metteniana)组织培养繁殖体系,优化其繁殖过程,通过小黑桫椤孢子萌发后的原叶体作为外植体,研究不同生长素和细胞分裂素对配子体增殖、分化及孢子体生根的影响。结果表明:配子体增殖,在1/2 MS+KT 0.3 mg/L+蔗糖30 g/L+琼脂6 g/L中增殖效果最好,增殖倍数为14.84倍,增殖后鲜重为1.51 g,直径为2.43 cm;配子体分化,在1/2 MS+KT 0.7 mg/L+蔗糖30 g/L+琼脂6 g/L中分化率最高,分化率为85.93%;孢子体生根,在1/2 MS+IBA 0.5 mg/L+蔗糖30 g/L+琼脂6 g/L中生根率最高,生根率达77.78%,平均生根数为1.42根。To preliminarily establish a tissue culture propagation system for Gymnosera metteniana and optimize its reproduction process,the prothalli derived from spore germination were used as explants to study the effects of different auxins and cytokinins on the proliferation and differentiation of gametophytes,as well as rooting of sporophytes.The results revealed that for gametophyte proliferation,the best result was obtained in 1/2 MS medium with 0.3 mg/L kinetin(KT),30 g/L sucrose,and 6 g/L agar,with a proliferation multiplier of 14.84 times.The fresh weight after proliferation was 1.51 g,and the diameter was 2.43 cm.The highest differentiation rate of the gametophyte was achieved in 1/2 MS medium with 0.7 mg/L KT,30 g/L sucrose,and 6 g/L agar,with a differentiation rate of 85.93%.The best rooting of sporophytes occurred in 1/2 MS medium with 0.5 mg/L indole-3-butyric acid(IBA),30 g/L sucrose,and 6 g/L agar,with the highest rooting rate of 77.78%and an average of 1.42 roots per sporophyte.
