摘  要:通过体外生化实验和细胞模型,研究牡丹皮提取物(Moutan Cortex Extract, MCE)的抗氧化和美白功效。实验表明,MCE对DPPH自由基、·OH自由基、ABTS^(+)自由基的清除率IC_(50)分别为(15.13±0.62),(560.4±12.8)和(51.91±0.70)μg/mL,对铁离子的还原能力随质量浓度的上升而增大。加入0.2 mg/mL MCE的HaCaT细胞在长波紫外线(UVA)照射后,MDA含量显著降低,对ABTS^(+)自由基的清除能力与空白对照组接近,能减少UVA照射引起的细胞氧化应激损伤。通过B16细胞实验评价美白效果,发现与0.1 mg/mL α-熊果苷相比,0.2 mg/mL MCE对B16细胞中黑色素的合成和酪氨酸酶活性的抑制效果更加突出。由此可知,MCE具有一定的抗氧化和美白功效。The antioxidant and whitening effects of the moutan cortex extract(MCE)were studied by biochemical experiments and cell models in vitro.The results showed that the IC_(50) values for scavenging DPPH radicals,hydroxyl radicals(·OH),and ABTS^(+)radicals by MCE were(15.13±0.62),(560.4±12.8)and(51.91±0.70)μg/mL,respectively.The reducing capacity of iron ion increased with the increase of the concentration.When HaCaT cells were treated with 0.2 mg/mL MCE and exposed to long-wave ultraviolet(UVA)light,the MDA content significantly decreased,and the scavenging ability of ABTS^(+) free radicals was similar to that of blank control group,which reduced the oxidative stress damage of cells caused by UVA irradiation.The whitening effect was evaluated using B16 cells,and it was found that at a concentration of 0.2 mg/mL,MCE exhibited a more pronounced inhibitory effect on melanin synthesis and tyrosinase activity in B16 cells compared to 0.1 mg/mLα-arbutin.These results indicate that MCE possesses certain antioxidant and whitening properties.
