云南重楼侧根的化学成分研究

摘  要:目的:研究云南重楼(Paris polyphylla var.Yunnanensis)侧根中的75%乙醇提取物的化学成分。方法:采用YWD-3F大孔吸附树脂、硅胶层析柱、反相材料(Rp-18)、葡聚糖凝胶(Sephadex LH-20)和制备以及半制备HPLC等多种材料和分离手段,结合NMR、MS、UV、IR等现代波谱技术和化学方法进行结构鉴定。结果:从云南重楼非药用部位侧根中分离得到10个化合物,分别鉴定为重楼皂苷VI(1)、偏诺皂苷元(2)、(3β,17α,25R)-spirost-5-en-3,17-diol-3-O-α-L-arabinofuranosyl-(1→4)-β-D-glucopyranoside[17-羟基-3-O-α-L-阿拉伯糖基-(1→4)-β-D-葡萄吡喃糖苷](3)、(3β,17α,25R)-spirost-5-en-3,17-diol-3-O-α-D-xylopyranosyl-(1→5)α-L-arabinofuranosyl-(1→4)-β-D-glucopyranoside[17-羟基-3-O-α-D-木糖基-(1→5)-α-L-阿拉伯糖基-(1→4)-β-D-葡萄吡喃糖苷](4)、重楼皂苷V(polyphyllin V)(5)、diosgenin-3-O-α-L-rhamnopyranosyl-(1→2)-[α-L-rhamnopyranosyl-(1→4)-α-L-rhamnopyranosyl-(1→4)]-β-D-glucopyranoside(6)、3-羟基-(3β)-乌索-12-烯-28-酸乌索酸(Ursolic acid)(7)、三裂鼠尾草素(salvigenin)(8)、3Ac-O-5β-蜕皮激素(3Ac-O-5β-ecdysterone)(9)、4-ethenyl-1-O-β-D-apiosyl-(1→6)-β-D-glucopyranoside(10)。结论:非药用部位须根和药用部位根茎部分具有相同的化学成分,从药用资源的开发利用角度来考虑,云南重楼须根部分具有一定的利用价值。Objective:To study the chemical constituents and it’s anti-acetylcholinesterase activity from 75%ethanol extract of the lateral roots of Paris polyphylla Smith var.yunnanensis(Franch.)Hand.-Mazz.Methods:The chemical constituents were isolated and purified by various separation materials and methods over macroporous resin,silica gelcolumn,reverse phase column(Rp-18),Sephadex LH-20,semi-preparative HPLC,etc.,combinedwith comprehensive spectroscopic technique such as^(1)H-NMR,^(13)C-NMR,MS,UV,IR,etc.Results:Ten chemical compound were isolated from the lateral roots of Parispolyphylla var.yunnanensis and identified as:pennogenin3-O-α-L-rhamnopyranosyl(1→2)-β-D-glucopyranoside(1),pennogenin(2),(3β,17α,25R)-spirost-5-en-3,17-diol-3-O-α-L-arabinofuranosyl-(1→4)-β-D-glucopyranoside(3),(3β,17α,25R)-spirost-5-en-3,17-diol-3-O-α-D-xylopyranosyl-(1→5)α-L-arabinofuranosyl-(1→4)-β-D-glucopyranoside(4),polyphyllin V(5),diosgenin-3-O-α-L-rhamnopyranosyl-(1→2)-[α-L-rhamnopyranosyl-(1→4)-α-L-rhamnopyranosyl-(1→4)]-β-D-glucopyranoside(6),Ursolic acid(7),salvigenin(8),3Ac-O-5β-ecdysterone(9),4-ethenyl-1-O-β-D-apiosyl-(1→6)-β-D-glucopyranoside(10),respectively.Conclusion:The essential parts,such as the lateral roots,and the medicinal parts,such as the rhizomes,have the same chemical composition.From the perspective of the development and utilization of medicinal resources,thethe lateral roots of Paris polyphylla Smith var.yunnanensis(Franch.)Hand.-Mazz.have certain utilization value.
