摘  要:目的:通过检测不同配比黄连-吴茱萸药对对成熟的小鼠棕色脂肪细胞中解偶联蛋白(UCP1)基因表达和蛋白表达的影响,探索中药寒热药性的分子生物学本质。方法:诱导小鼠原代前棕色脂肪细胞,待分化为成熟的棕色脂肪细胞,油红O染色鉴定并观察细胞形态变化。用荧光定量PCR技术和Western blot技术研究不同配比黄连-吴茱萸药对(左金丸、反左金丸、痞满方)以及单味中药及单体(黄连、吴茱萸、黄连素、吴茱萸碱)对成熟棕色脂肪细胞中UCP1在基因和蛋白表达层面的影响。结果:不同配比的“连-萸”药对会使UCP1、PGC-1α蛋白和基因相对表达量发生变化,偏寒性“连-萸”药对(左金丸、痞满方、黄连)会使UCP1、PGC-1α蛋白和基因相对表达量下降(P<0.05),偏热性“连-萸”药对(反左金丸、吴茱萸)会使UCP1、PGC-1α蛋白和基因相对表达量上升(P<0.05)。结论:具有寒热偏性的中药药对可以通过调节UCP1、PGC-1α蛋白和基因相对表达来影响能量代谢。在分子水平上解释中药寒热药性的科学本质。Objectives To explore the molecular biological nature of cold-heat properties of traditional Chinese medicine by detecting the effects of different ratios of Coptis and Evodia officinaris on UCP1 gene and protein expression in mature mouse brown adipocytes.Methods The pre-primary brown fat cells of mice were induced to differentiate into mature brown fat cells,and the morphological changes of the cells were identified by oil red O staining.Quantitative PCR and Western blot were used to study the effects of different ratios of Coptis and Evodia on UCP1 gene and protein expression in mature brown adipocytes,including Zuojin pills,anti-Zuojin pills,and Piman fang,as well as single Chinese herbs and monomer(Coptis,Evodia,berberine,Evodia).Results The relative expression levels of UCP1 and PGC-1αprotein and gene were changed by different proportions of“Coptis and Evodia”drug pairs,while the relative expression levels of UCP1 and PGC-1αprotein and gene were decreased by cold“Coptis and Evodia”drug pairs(Zuojin pills,Piman fang and Coptis)(P<0.05).The relative expression levels of UCP1 and PGC-1αprotein and gene were increased by the heat-related“Coptis and Evodia”pairs(anti-zuojin pill and Evodia)(P<0.05).Conclusion Chinese medicine pairs with cold-heat bias can affect energy metabolism by regulating UCP1,PGC-1αprotein and gene relative expression.The scientific nature of explaining the medicinal properties of cold and heat of traditional Chinese medicine at molecular level.
