摘 要:短节间相关序列(SHI-related sequence,SRS)基因家族编码一类植物特有的锌指蛋白转录因子,在调节植物生长发育和响应逆境胁迫中发挥重要作用。为探究烟草SRS基因家族功能,本研究从烟草基因组数据库中鉴定了18个NtSRS基因,命名为NtSRS1~NtSRS18。利用生物信息学方法,分析了该家族基因的蛋白质特征、亚细胞定位、系统进化关系、基因结构、启动子结构及其组织表达模式和对干旱、盐、低温胁迫的响应模式。结果表明:NtSRS蛋白都含有DUF702保守结构域,大部分NtSRS蛋白定位于细胞核上。NtSRS基因家族聚类为5个亚家族,且同一亚家族成员的基因和蛋白质结构均具有较高的保守性;启动子结构分析表明,NtSRS基因家族成员的启动子区存在大量激素和逆境响应元件。NtSRS基因在不同组织器官中的表达量存在较大差异,大多数基因主要在花芽和茎中表达。NtSRS3和NtSRS16基因对干旱、盐和低温胁迫都有响应。本研究为烟草SRS基因家族的生物学功能研究奠定了基础,为烟草种质资源改良提供了理论依据。The SHI-related sequence(SRS)gene family encodes a class of plant-specific zinc finger protein transcription factors that play crucial roles in regulating plant growth,development and stress responses.To explore the function of the SRS gene family in tobacco,18 NtSRS genes were identified from the tobacco genome database and designated as NtSRS1 to NtSRS18.Bioinformatics analyses were conducted to investigate their protein characteristics,subcellular localization,phylogenetic relationships,gene structures,and promoter elements.Additionally,expression patterns of NtSRS family across different tissues and organs,as well as their responses to drought,salt,and low temperature stress,were analyzed using qRT-PCR.Results revealed that NtSRS proteins contained the conserved DUF702 domain,with most localized in the nucleus.Phylogenetic analysis clustered the NtSRS family into 5 subfamilies,with members of the same subfamily exhibiting highly conserved gene and protein structures.Promoter analysis identified numerous hormone-responsive elements and stress-responsive elements within the regulatory regions of NtSRS genes.Expression profiling showed that NtSRS genes exhibited significant tissue-specific expression,with most highly expressed in flower buds and stems.Notably,NtSRS3 and NtSRS16 were strongly induced under drought,salt,and low temperature stress conditions.Overall,this study provides a comprehensive characterization of the tobacco SRS gene family,offering a theoretical basis for the genetic improvement of tobacco germplasm resources.
