摘 要:目的研究跑台运动对2型糖尿病大鼠认知功能障碍和突触功能的影响及其潜在机制。方法雄性Sprague-Dawley(SD)大鼠45只,随机分为对照组(Control)、糖尿病组(T2DM)、运动干预组(RE)。采用长期高脂饮食加一次性小剂量链脲佐霉素(STZ)尾静脉注射来制作T2DM大鼠模型,Control大鼠采用普通饮食加一次等剂量生理盐水尾静脉注射作为对照,RE大鼠在造模期间同时进行4个月的跑台运动,并使用水迷宫实验检测认知功能,电生理检测海马突触可塑性,免疫印迹法(Western blotting)检测海马中突触相关蛋白表达水平,免疫组化检测海马中淀粉样蛋白-β(Aβ)的表达。结果跑台运动干预可有效改善T2DM大鼠的认知功能障碍和海马神经元细胞突触可塑性,增加海马中突触相关蛋白的表达水平,并降低海马Aβ表达。结论跑台运动能够有效改善T2DM大鼠认知功能障碍和突触功能损伤。Objective To study the effects of running treadmill exercise on cognitive dysfunction and synaptic function in type 2 diabetic rats and its underlying mechanisms.Methods Forty-five male Sprague-Dawley(SD)rats were randomly divided into the control group(Control group),the type 2 diabetes mellitus group(T2DM group),and the exercise intervention group(RE group).A model of T2DM rats was established by long-term high-fat diet combined with a single low-dose streptozotocin(STZ)injection via the tail vein.The rats in the Control group were given a normal diet and a single injection of an equal dose of normal saline via the tail vein as a control.The rats in the RE group underwent running treadmill exercise for 4 months during the modeling period.The Morris water maze test was used to detect cognitive function,electrophysiological examination was used to detect hippocampal synaptic plasticity,Western blotting was used to detect the expression levels of synaptic-related proteins in the hippocampus,and immunohistochemistry was used to detect the expression of amyloid-β(Aβ)in the hippocampus.Results Running treadmill exercise intervention effectively improved the cognitive dysfunction and synaptic plasticity of hippocampal neuron cells in T2DM rats,increased the expression levels of synaptic-related proteins in the hippocampus,and reduced the expression of Aβin the hippocampus.Conclusion Running treadmill exercise can effectively improve cognitive dysfunction and synaptic function damage in T2DM rats.
