光谱法和色谱法对原花青素含量测试的比对研究

摘  要:本文通过线性相关性、灵敏度、精密度和抗干扰能力试验,比较了采用分光光度法和高效液相色谱法检测原花青素的优劣。结果表明:分光光度法的线性相关系数为0.9994,高效液相色谱法的线性相关系数为0.9998,连续重复进样分光光度法的RSD为2.9%,高效液相色谱法为1.9%,线性相关性、精密度两种仪器分析方法都能满足测试要求;灵敏度测试中高效液相色谱法优于分光光度法,高效液相色谱法检出限为2.0 mg/L,分光光度法检出限为5.0 mg/L;对于一些水解反应后存在颜色干扰的样品,高效液相色谱法定性结果更准确。In this paper,through experiments on linear correlation,sensitivity,precision and anti-interference ability,the advantages and disadvantages of spectrophotometry and high-performance liquid chromatography in the detection of proanthocyanidins were compared.The linear correlation coefficient of spectrophotometry is 0.994,and that of high-performance liquid chromatography is 0.998.The relative standard deviation(RSD)of continuous repeated injections by spectrophotometry is 2.9%,while that of high-performance liquid chromatography is 1.9%.Both instrumental analysis methods of linear correlation and precision can meet the testing requirements.In the sensitivity test,HPLC was superior to spectrophotometry.The detection limits of HPLC were 2.0 mg/L,and those of spectrophotometry were 5.0 mg/L.For some samples with color interference after hydrolysis reac-tions,the qualitative results obtained by high-performance liquid chromatography are more accurate.
