摘 要:目的 评价白藜芦醇对糖尿病心肌病小鼠心肌细胞铁死亡的影响。方法 非随机选取2023年1—2月徐州医科大学附属邳州医院实验室80只C57BL/6小鼠,高脂肪饲养,构建糖尿病心肌病模型,按随机数字表法分为空白对照组(20只)、观察组(60只),观察组根据白藜芦醇干预浓度0.5 mg/100 g、1.5 mg/100 g、3 mg/100 g分为3个亚组(A组、B组、C组),每组20只。比较各组心肌损伤相关标志物、心肌组织铁含量、细胞铁死亡情况。结果 实验7 d后,A组、B组、C组乳酸脱氢酶、肌酸激酶同工酶水平均较对照组低,其中C组最低,差异均有统计学意义(P均<0.05)。A组、B组、C组心肌组织铁含量分别为(30.33±2.36)μmol/g、(26.22±2.11)μmol/g、(20.31±1.98)μmol/g,均少于对照组的(38.54±2.62)μmol/g,差异有统计学意义(F=225.487;P<0.05)。A组、B组、C组长链脂酰辅酶A合成酶4表达低于对照组,其中C组表达最低,差异均有统计学意义(P均<0.05)。A组、B组、C组谷胱甘肽过氧化物酶4、溶质载体家族7成员11表达高于对照组,其中C组表达最高,差异均有统计学意义(P均<0.05)。结论 3 mg/100 g浓度白藜芦醇可有效抑制糖尿病心肌病小鼠的心肌细胞铁死亡,改善心肌细胞损伤情况。Objective To evaluate the effect of resveratrol on iron death in cardiomyocytes in mice with diabetic car⁃diomyopathy.Methods Eighty C57BL/6 mice were non-randomly selected from the laboratory of Pizhou Hospital Af⁃filiated to Pizhou Hospital of Xuzhou Medical University from January to February 2023,high-fat feeding,construct⁃ing diabetic cardiomyopathy model,and divided into a blank control group(twenty mice)and an observation group(sixty mice)according to the randomized numerical table method.The observation group was divided into three sub⁃groups(group A,group B,group C)according to the resveratrol intervention concentration of 0.5 mg/100 g,1.5 mg/100 g,3 mg/100 g,with twenty mice in each group.Myocardial injury-related markers,myocardial tissue iron content,and cellular iron death were compared in each group.Results After 7 d of the experiment,the levels of L-lactate dehydro⁃genase and cre-atine kinase isoenzyme in group A,group B,and group C were lower than those in the control group,with group C having the lowest level,and the differences were statistically significant(all P<0.05).The iron content of myocardial tissues in group A,group B,and group C were(30.33±2.36)μmol/g,(26.22±2.11)μmol/g,and(20.31±1.98)μmol/g,all of which were less than(38.54±2.62)μmol/g in the control group,and the difference was statistically significant(F=225.487;P<0.05).The expression of acyl-CoA synthetase long chain family member 4 in group A,group B,and group C were lower than that in the control group,and the expression in group C was the lowest,and the differences were statistically significant(all P<0.05).The expression of glu-tathione peroxidase 4 and solute carrier family 7 member 11 in group A,group B,group C were higher than those of the control group,and the expression in group C was the highest,and the differences were statistically significant(all P<0.05).Conclusions Resveratrol at a concentration of 3 mg/100 g effectively inhibited cardiomyocyte iron death and ameliorated cardiomyocyte injury in mice
