摘 要:为探究抗生素抗性基因(ARGs)在可生物降解塑料PBAT/PLA共聚物和传统塑料聚丙烯表面的赋存规律,以评估可生物降解塑料在水生环境中的生态风险,该文在上海黄浦江支流进行为期2个月的原位暴露实验,采用宏基因组测序分析塑料表面微生物群落和ARGs变化规律。结果表明,PBAT/PLA表面ARGs的总含量为199~340 RPKM,且更易携带RanA、qacEdelta1和msbA等多重耐药基因,其移动遗传元件(MGEs)丰度高达4866~7904 RPKM,以转座子和插入序列为主。在PBAT/PLA表面ARGs的潜在宿主属有17个,且脱硫弧菌和假芽孢杆菌等均与sul2和mexK等多种ARGs呈显著正相关。PBAT/PLA塑料表面高MGEs总丰度、以抗生素失活为主的多种抗性机制并存以及核心潜在宿主与ARGs呈现的“一对多”关联模式共同驱动了ARGs的富集,加剧耐药性的传播风险。To assess ecological risks of biodegradable plastics in aquatic environments,a two-month in-situ exposure experiment was conducted in a Shanghai Huangpu River tributary,analyzing microbial communities and antibiotic resistance genes(ARGs)variation patterns on PBAT/PLA copolymers and polypropylene plastics using metagenomic sequencing.The total ARGs in PBAT/PLA ranged from 199 to 340 RPKM,and it was more prone to harbor multiple antibiotic resistance genes such as RanA,qacEdelta1,and msbA.The abundance of mobile genetic elements(MGEs)in PBAT/PLA was from 4866 RPKM to 7904 RPKM,dominated by transposons and insertion sequences.There were 17 potential host genera of ARGs in PBAT/PLA,with Desulfovibrio and Fictibacillus barbaricus showing significant positive correlations with various ARGs including sul2 and mexK.The high total abundance of MGEs on the surface of PBAT/PLA plastics,the coexistence of multiple resistance mechanisms mainly based on antibiotic inactivation,and the“one-to-many”correlation pattern between core potential hosts and ARGs jointly drive the enrichment of ARGs and exacerbate the risk of the spread of drug resistance.
