摘 要:蓝宝石眼睛是尼里拉菲水牛的重要品种特征。为了研究水牛不同颜色眼睛的基因表达模式,本研究对黑色瞳色的摩拉水牛和蓝色瞳色的尼里拉菲水牛的眼睛进行RNA测序(RNA sequencing,RNA-seq),并以|log2FC|≥2且P<0.05为筛选标准,对差异表达基因进行GO和KEGG富集分析,进一步对通路上的基因进行蛋白质互作分析。结果显示,有1708个基因在摩拉水牛眼睛和尼里拉菲水牛眼睛之间有差异表达。富集分析显示,差异表达基因主要富集在黑色素生成、酪氨酸代谢、半胱氨酸代谢和光传导等通路。本研究还挖掘出多个与眼睛光传导和黑色素生成相关的关键差异基因,确定了KIT、MGST1、GNGT1和TYRP1等在功能网络中高度互联的核心基因。本研究为尼里拉菲水牛蓝色瞳色形成的分子调控机制提供了参考,扩展了对水牛眼睛中黑色素沉积的生物学认识。The sapphire eyes are a distinctive trait of the Nili-Ravi buffalo breed.To investigate the gene expression patterns associated with different eye colors in buffaloes,we performed RNA sequencing(RNA-seq)on the eyes of both black-eyed Murrah buffaloes and blue-eyed Nili-Ravi buffaloes.Using|log_2FC|≥2 and P<0.05 as selection criteria,we conducted differential expression gene analysis,followed by GO and KEGG enrichment analyses.Additionally,protein-protein interaction analysis on the genes within these pathways was performed.The results identified 1708 differentially expressed genes between the eyes of Murrah and Nili-Ravi buffaloes.Enrichment analysis revealed that these genes were mainly enriched in melaninbiosynthesis,tyrosine metabolism,cysteine metabolism,and phototransduction.We also identified several key differential genes related to eye phototransduction and melanin biosynthesis,including KIT,MGST1,GNGT1,and TYRP1,which were highly interconnected within functional networks.This study provides insights into the molecular mechanisms underlying the formation of blue eye color in Nili-Ravi buffaloes and enhances our understanding of melanin deposition in buffalo eyes.
