摘  要:目的探究复方菝葜丹参擦剂(compound baqia danshen liniment,BQDS)通过调控Janus激酶(janus kinase-signal transducer,JAK)/信号转导及转录激活因子(signal transducer and activator of transcription,STAT)信号通路对白癜风症状的影响。方法将C57BL/6小鼠随机分为对照组(Control)、模型组(Model)、阳性药物组(复方卡力孜然酊)、BQDS组及BQDS+RO8191组(JAK激活剂),在小鼠脱毛区连续21 d涂40%的莫诺苯宗乳膏,每次0.5 mL、2次/d,建立白癜风小鼠模型;BQDS组涂抹BQDS,BQDS+RO8191组涂抹BQDS并腹腔注射2 mg/kg的RO8191,Control组和Model组涂抹等体积蒸馏水,治疗30 d;治疗结束后评估小鼠皮肤毛发脱色情况;采用苏木精伊红(HE)和Masson-Fontana染色观察实验区域皮肤含黑色素的毛囊数和基底层黑素细胞数,酶联免疫吸附(ELISA)法检测血清单胺氧化酶(monoamine oxidase,MAO)、酪氨酸酶(tyrosinase,TYR)、α-黑色素细胞刺激素(alpha-melanocyte stimulating hormone,α-MSH)、肿瘤坏死因子-α(tumor necrosis factor-alpha,TNF-α)及白细胞介素-6(Interleukin-6,IL-6)水平;试剂盒检测皮肤组织丙二醛(malondialdehyde content,MDA)含量、超氧化物歧化酶(superoxide dismutase,SOD)及谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性;蛋白免疫印迹法检测小眼畸形相关转录因子(microphthalmia-associated transcription factor,MITF)、酪氨酸酶相关蛋白-1(tyrosinase-related protein 1,TRP-1)、酪氨酸酶相关蛋白-2(tyrosinase-related protein 2,TRP-2)及JAK-STAT信号通路蛋白表达。结果与Control组相比,Model组小鼠脱色评分、MAO、TNF-α和IL-6水平、MDA含量、p-JAK/JAK和p-STAT/STAT蛋白表达水平升高,黑色素毛囊数量和基底层黑素细胞数量减少、TYR和α-MSH水平、SOD和GSH-Px活性、MITF、TRP-1和TRP-2蛋白表达水平降低(P<0.05);与Model组相比,阳性药物组和BQDS组小鼠脱色评分、MAO、TNF-α和IL-6水平、MDA含量、p-JAK/JAK和p-STAT/STAT蛋白表达水平降低,黑色素Objective To investigate whether compound Baqia Danshen liniment(BQDS)can improve symptoms of vitiligo in mice by regulating the Janus kinase-signal transducer(JAK)/signal transducer and activator of transcription(STAT)signaling pathway.Methods 57BL/6 mice were randomly divided into control group,model group,positive drug(compound Kaliziran tincture)group,BQDS group,and BQDS+RO8191(JAK activator)group.The mouse model of vitiligo was established by continuously applying 40%monobenzone cream(0.5 mL each time,twice a day)to the depilated area of the mice for 21 consecutive days.The BQDS group was treated with BQDS.The BQDS+RO8191 group was treated with BQDS and intraperitoneal injection of 2 mg/kg RO8191.Control group and model group were treated with distilled water of equal volume.The treatment lasted for 30 days.When the treatment was completed,the depigmentation of mice skin and hair was evaluated.Hematoxylin eosin staining and Masson fontana staining were applied to detect the number of hair follicles containing melanin and the number of melanocytes in the basal layer of the skin in the experimental area;Enzyme-linked immunosorbent assay to detect the levels of serum monoamine oxidase(MAO),tyrosinase(TYR),alpha melanocyte stimulating hormone(α-MSH),TNF-α,and IL-6;and the reagent kit to detect the malondialdehyde content(MDA),superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px)activities in skin tissues;Western blot to detect the expression of microphthalmia-associated transcription factor(MITF),tyrosinase-related protein 1(TRP-1),TRP-2,and JAK-STAT signaling pathway proteins.Results Compared with control group,decolorization score,MAO,TNF-α,and IL-6 levels,MDA content,p-JAK/JAK,and p-STAT/STAT protein expression levels were reduced in model group.The number of melanin hair follicles and basal melanocytes decreased,as well as TYR andα-MSH levels,SOD,and GSH-Px activities,MITF,TRP-1,and TRP-2 protein expression levels decreased(P<0.05).Compared with Model group,decolorization score,MAO,TNF-α,and IL-6
