秋水仙素诱导阳光玫瑰葡萄多倍体的效果研究

摘  要:以阳光玫瑰葡萄茎段为试材,利用0.2%、0.4%、0.8%、1.2%的秋水仙素溶液,对刻芽的枝段采用棉浸法和浸泡法处理不同时间,以获得四倍体种质。结果表明,诱变液以0.4%秋水仙素2000 mL+1%二甲基亚砜200 mL浸泡葡萄枝段6 d的诱变效果最好,诱变致死率和变异率分别是50.00%和6.60%,共获得阳光玫瑰四倍体植株12株和混倍体2株;棉浸法以1.2%秋水仙素200 mL+1%二甲基亚砜20 mL棉浸葡萄枝段6 d的诱变效果最好,诱变致死率和变异率分别是25.00%和6.00%。The shoot segments of Shine Muscat grapes were used as materials,and 0.2%,0.4%,0.8%,and 1.2%colchicine solutions were applied to treat the shoot with different times by the cotton immersion and soaking methods,the study was aimed to obtain the tetraploid grapes.The results showed that soaking grape branch segments in 0.4%colchicine(2000 mL)and 1%dimethyl sulfoxide(200 mL)for 6 days resulted in the greatest mutagenic effect,with a lethal rate of 50.00%and a mutation rate of 6.60%,respectively.A total of 12 tetraploid and 2 mixed polyploid plants of the Sunshine Rose were obtained;The cotton soaking method with 1.2%colchicine 200 mL+1%dimethyl sulfoxide 20 mL cotton soaked grape branch segments for 6 days showed the greatest mutagenic effect,with a lethal and mutation rate of 25.00%and 6.00%,respectively.
